[origin: WO8203077A1] In the method for the synthesis of a polypeptide chain on an insoluble solid support wherein an amino acid is introduced into a reactor packed with an insoluble solid support containing substituent groups reactive with said amino acid, coupling said amino acid to said support by condensation reaction with said reactive substituent groups, introducing a second similar or dissimilar amino acid into said reactor and coupling said second amino acid to said first amino acid and repeating the process until the desired polypeptide is obtained, the improvement wherein a pressurized flow system is used to obtain rapid reactions and quantitative yields of the desired products.

Procede de synthese d'une chaine de polypeptide sur un support solide insoluble, dans lequel un acide amine est introduit dans un reacteur rempli d'un support solide insoluble contenant des groupes substituants reagissant avec cet acide amine, le procede consistant a coupler cet acide amine au support par une reaction de condensation avec les groupes de substituants reactifs, a introduire un deuxieme acide amine analogue ou different dans le reacteur et a coupler le deuxieme acide amine au premier acide amine et a repeter le procede jusqu'a obtention du polypeptide desire, l'amelioration etant constituee par l'utilisation d'un systeme d'ecoulement sous pression permettant d'obtenir des reactions rapides et des rendements de productions eleves des produits desires.
