[origin: WO8701730A1] A stable, single-stranded nucleoprotein filament adapted to complex specifically and stably with a target duplex DNA having a selected base sequence. The filament is composed of a single-stranded DNA probe having a region of homology with the target base sequence, and RecA protein bound stably to the DNA probe by adenosine 5'-( gamma -thio)triphosphate. The filament is useful in a novel system and method for enriching target duplex DNA which contains a region homologous to the probe sequence, for blocking selected restriction endonuclease sites in the target DNA, and in other DNA methodologies in which stable rapid triple-strand synaptic formation in duplex DNA can be exploited.

Un filament de nucléoprotéines stables monobrin est adapté pour produire un complexe de manière spécifique et stable avec un ADN duplex cible ayant une séquence de base sélectionnée. Le filament se compose d'un agent d'investigation d'ADN monobrin ayant une région d'homologie avec la séquence de base cible, et une protéine RecA liée de manière stable à l'agent d'investigation d'ADN par de l'adénosine 5'-(gamma-thio)triphosphate. Le filament est utile dans un nouveau système et un procédé pour enrichir d'ADN duplex cible qui contient une région homologue à la séquence d'investigation pour bloquer des sites d'endonucléase de restriction sélectionnés dans l'ADN cible, ainsi que dans d'autres méthodologies relatives à l'ADN dans lesquellles la formation synaptique triple brin stable, rapide dans l'ADN duplex peutêtre exploitée.
