Abstract (en)
The process entails the medium being exchanged by ultrafiltration. The culture medium is first removed from the reactor through a microporous membrane to give a cell-free medium. This medium is passed to an ultrafiltration membrane for ultrafiltration. The filtrate containing low molecular weight components and metabolic products is discarded, and the concentrate is returned to the reactor. Fresh medium corresponding to the amount of filtrate is fed into the reactor from a medium reservoir.

Die vorliegende Erfindung betrifft ein Verfahren zur Kultivierung von Säugerzellen in Suspensionskultur wobei der Mediumaustausch durch Ultrafiltration erfolgt. Das Kulturmedium wird zuerst über eie mikroporöse Membran aus dem Reaktor abgezogen um ein zellfreies Medium zu erhalten. Dieses Medium wird einer Ultrafiltrationsmembran zugeführt und ultrafiltriert. Das die niedermolekularen Bestandteile und Stoffwechselprodukte enthaltende Filtrat wird verworfen und das Konzentrat dem Reaktor wieder zugeführt. Aus einem Mediumreservoir wird dem Reaktor entsprechend der Menge an Filtrat frisches Medium zugeführt.
