[origin: WO8703911A1] Diagnostic assay methods for detecting the presence of a target nucleotide sequence (TNS) in a biological sample. A reagent complex for use in the first method is also provided which includes a labelled probe (LP) having a target binding region (TBR) and a second polynucleotide (SP) which binds to the probe in at least a portion of the target binding region. The complex is contacted with sample (G) and target displaces the second polynucleotide on the complex. Intact complex is then separated from probe/target hybrids, and the hybrids are detected. The second method involves capturing of the labeled strand after displacement from the probe which binds to the target.
