[origin: WO8805823A2] Mycobacterium tuberculosis genes encoding five immunologically relevant proteins have been isolated by systematically screening a lambda gt11 recombinant DNA expression library with a collection of murine monoclonal antibodies directed against protein antigens of this pathogen. One of the M. tuberculosis antigens, a 65kD protein, has been shown to have determinants common to M. tuberculosis and M. leprae. In addition, genes encoding proteins of other mycobacteria (M. africanum, M. smegmatis, M. bovis BCG and M. avium) have been isolated. Isolation and characterization of genes encoding major protein antigens of M. tuberculosis make it possible to develop reagents useful in the diagnosis, prevention and treatment of tuberculosis. They can be used, for example, in the development of skin tests, serodiagnostic tests and vaccines specific for tuberculosis.

Des gènes de Mycobacterium tuberculosis codant cinq protéines à effet immunologique ont été isolés en criblant systématiquement une librairie d'expressions d'ADN recombinant lambda gt11 avec une collection d'anticorps monoclonaux murins dirigés contre des antigènes de protéines de ce pathogène. L'un des antigènes M. tuberculosis, une protéine 65kD possède des déterminants communs à M. tuberculosis et M. leprae. De plus, des gènes codant des protéines d'autres mycobactéries M. africanum, M. smegmatis, M. bovis BCG et M. avium ont été isolés. L'isolement et la caractérisation de gènes codant des antigènes de protéines majeures de M. tuberculosis permet de développer des réactifs utiles pour les diagnostics, la prévention et le traitement de la tuberculose. Ils peuvent être utilisés par exemple dans le développement de tests épidermiques, de tests sérodiagnostiques et de vaccins contre la tuberculose.
