[origin: EP0421469A2] A method for separating a nuleic acid comprises conducting hybridization reaction of a target nucleic acid with a probe nucleic acid in a mixture of a reaction solution, and extracting the resulting hybrid former composed of the separated target nucleic acid and the probe nucleic acid from the mixture of the reaction solution: said method comprising the steps of a) introducing an immobilizing substance, which reacts specifically with a carrier, only into the hybrid former by extension reaction; and b) separating the hybrid former having the immobilizing substance by bonding the immobilized substance to a carrier from a non-hybrid former in the reaction solution. A method for detecting the nucleic acid comprises utilizing a labelling substance in the separated hybrid former prepared by the method.
