[origin: WO9110747A1] An immunoassay for determining a first antigen (Ag1) and/or an antibody (Ab2) to a second antigen (Ag2) in a sample, which immunoassay comprises the steps: (a) contacting the sample with a solid support on which Ag2 and an antibody (Ab1) to Ag1 have been coated so that sample Ag1 and sample Ab2, if present, will become bound to Ab1 and Ag2, respectively; (b) adding Ag2 in the form of a detecting reagent; the addition being performed under conditions allowing the formation of an immune complex between added Ag2 and sample Ab2 bound to the solid support in step (a); (c) adding Ab1 in the form of a detecting reagent; the addition being performed under conditions allowing the formation of an immune complex between added Ab1 and sample Ag1 bound to the solid support in step (a); (d) measuring the amount of immune complex formed in step (b) and (c) and taking the amount as an indication of the amount of Ag1 and/or Ab2 in the sample. All or part of the antigenic entitie(s) in Ag2 coated on the solid support and/or Ag2 used as the detecting reagent are derived from different sources. In an alternative mode Ag2 and Ab1 in the detecting reagent is equipped with biotin so that the immune complexes formed in steps (b) and (c) can be measured by the use of a biotin binding substance.

Immunoanalyse pour déterminer un premier antigène (Ag1) et/ou un anticorps (Ab2) d'un deuxième antigène (Ag2) dans un échantillon. L'immunoanalyse comprend les étapes suivantes: (a) la mise en contact de l'échantillon avec un support solide sur lequel ont été déposés le deuxième antigène Ag2 et un anticorps (Ab1) du premier antigène (Ag1) de sorte que l'échantillon Ag1 et l'échantillon Ab2, éventuellement, vont se lier respectivement à l'Ab1 et à l'Ag2; (b) l'adjonction d'Ag2 sous forme d'un réactif de détection; l'adjonction s'effectuant dans des conditions permettant la formation d'un complexe immun entre l'Ag2 ajouté et l'échantillon Ab2 lié au support solide dans l'étape (a); (c) l'adjonction d'Ab1 sous forme d'un réactif de détection; l'adjonction étant réalisée dans des conditions permettant la formation d'un complexe immun entre l'Ab1 ajouté et l'échantillon Ag1 lié au support solide dans l'étape (a); (d) la mesure de la quantité de complexe immun formé dans l'étape (b) et (c), et la prise en compte de cette quantité comme une indication de la quantité d'Ag1 et/ou d'Ab2 présente dans l'échantillon. La totalité ou une partie de la ou des entité(s) antigénique(s) située(s) dans l'Ag2 déposé sur le support solide et/ou l'Ag2 utilisé comme réactif de détection proviennent de sources différentes.
