[origin: WO9110747A1] An immunoassay for determining a first antigen (Ag1) and/or an antibody (Ab2) to a second antigen (Ag2) in a sample, which immunoassay comprises the steps: (a) contacting the sample with a solid support on which Ag2 and an antibody (Ab1) to Ag1 have been coated so that sample Ag1 and sample Ab2, if present, will become bound to Ab1 and Ag2, respectively; (b) adding Ag2 in the form of a detecting reagent; the addition being performed under conditions allowing the formation of an immune complex between added Ag2 and sample Ab2 bound to the solid support in step (a); (c) adding Ab1 in the form of a detecting reagent; the addition being performed under conditions allowing the formation of an immune complex between added Ab1 and sample Ag1 bound to the solid support in step (a); (d) measuring the amount of immune complex formed in step (b) and (c) and taking the amount as an indication of the amount of Ag1 and/or Ab2 in the sample. All or part of the antigenic entitie(s) in Ag2 coated on the solid support and/or Ag2 used as the detecting reagent are derived from different sources. In an alternative mode Ag2 and Ab1 in the detecting reagent is equipped with biotin so that the immune complexes formed in steps (b) and (c) can be measured by the use of a biotin binding substance.
