[origin: WO9113166A1] Disclosed is an antibody construct with enhanced binding activity specific for an epitope on an antigen. The construct includes an immunoglobulin binding region of predetermined specificity and having plural hypervariable regions homologous with a hypervariable region of a native antibody specific for the epitope. The construct further includes an immunoglobulin constant region consisting essentially of a CH1 domain, a hinge, and a CH3 domain and omitting the CH2 domain normally present in a native antibody. The immunoglobulin constant region enhances the binding activity of the construct at least two fold in comparison with the binding activity of a native antibody having the same specifity.

Construction d'anticorps ayant une activité de liaison améliorée spécifique à un déterminant antigénique sur un antigène. Ladite construction comprend une région de liaison de l'immunoglobuline de spécificité prédéterminée et ayant plusieurs régions hypervariables identiques à une région hypervariable d'un anticorps non dénaturé spécifique audit déterminant antigénique. Ladite construction comprend en outre une région de constance de l'immunoglobuline consistant essentiellement en un domaine CH1, une charnière et un domaine CH3 et omettant le domaine CH2 normalement présent dans un anticorps non dénaturé. La région de constance de l'immunoglobuline améliore au moins deux fois l'activité de liaison en comparaison à l'activité de liaison d'un anticorps non dénaturé ayant la même spécificité.
