[origin: WO9217786A1] A method for detecting chitin-containing organisms such as fungi, yeast and insects in a sample attached to a solid phase, by detecting the presence of chitin by using enzymes which specifically bind chitin is disclosed. The method is preferably carried out by passing a sample which contains chitin through a membrane in order to capture and adhere the chitinous material on the membrane. A chitin-binding enzyme such as chitinase or lysozyme is added under conditions which allow it to bind to the chitin on the membrane. A signal generating compound is either directly conjugated to the chitinase (or lysozyme) or indirectly conjugated to either an anti-chitinase (or lysozyme) antibody or to an anti IgG antibody (which binds to the anti-chitinase antibody) to detect the chitin-chitinase complex. Reagents for the signal generating compound then are added to produce a detection signal which indicates the presence of the chitin. Kits which contain the necessary materials for carrying out the methods of the invention are also disclosed as are novel conjugates.

On décrit un procédé de détection d'organismes contenant de la chitine, tels que les champignons, la levure et les insectes, dans un échantillon fixé à une phase solide, et qui consiste à détecter la présence de la chitine au moyen d'enzymes qui se lient spécifiquement avec celle-ci. Le procédé consiste de préférence à faire passer l'échantillon contenant la chitine à travers une membrane afin de piéger et de faire adhérer la matière chitineuse à la membrane. Une enzyme qui se lie à la chitine, telle que la chitinase ou la lysozyme, est ajoutée dans des conditions qui lui permettent de se lier à la chitine sur la membrane. Un composé produisant un signal est soit directement conjugué avec la chitinase (ou lysozyme), soit indirectement conjugué avec un anticorps anti-chitinase (ou lysozyme) ou avec un anticorps anti-IgG (qui se lie avec l'anticorps anti-chitinase), afin de détecter le complexe chitine-chitinase. Des réactifs destinés au composé produisant un signal sont alors ajoutés afin de produire un signal de détection indiquant la présence de la chitine. Des kits contenant les matériaux nécessaires pour effectuer les procédés de l'invention sont aussi décrits, ainsi que de nouveaux conjugués.
