Abstract (en)

Appts. for detecting target reactant utilises a second reactant immobilised at selected locations on a support structure (212) in correspondence to a predetermined pattern, the immobilised reactant binding the target reactant when the two are brought into contact. A coherent light source (216) is used to illuminate the support structure and the predetermined pattern causes a holographic image to be generated when the reactants are bound. This image can be detected by detection means (215). Also claimed is a method for detecting the target reactant using the appts. PREFERRED - One of the target and immobilised reactants is an antibody or antigen, a nucleic acid, an enzyme, a protein, or a polypeptide. The support structure is well (210) having the immobilised reactant bound to its bottom surface or a layer of porous material thicker than the wavelength of the coherent light, the immobilised reactant being dispersed throughout the layer. The predetermined pattern compensates for any distorting interfaces (220) between support structure and coherent light source. The holographic image is an image of a small bright object.
