[origin: WO9639489A1] The present invention relates to a method of purifying mammalian hematopoietic stem cells from a bone marrow cell population. The present invention also relates to a method of separating cycling HSCs from quiescent HSCs in a purified mammalian HSC sample. Further, the invention relates to a method of identifying an agent which is a fluorescent, vital dye which is lipophilic for use in purifying mammalian HSCs. The present invention also relates to a method of transplanting bone marrow in a mammalian host, such as a human, comprising introducing into the host the purified HSCs described herein. The invention further relates to a method of in vivo administration of a protein comprising transfecting a purified HSC with a construct comprising DNA which encodes a protein of interest and then introducing the stem cell into the host where the protein of interest is expressed. The present invention also relates to hematopoietic stem cells purified using the methods described herein.
