[origin: DE19527160C1] Described is a method of determining the concentration of an enzyme by reacting the enzyme with a substrate solution and measuring the concentration of the dye thus formed by photometric measurement of the absorption of a test solution, the measurement being carried out in the presence of a second dye which is added to the test solution and which does not interfere with the measurement of the concentration of the dye formed by the enzyme. For the determination of alkaline phosphatase from a biological sample or from an enzyme immuno-assay, the preferred dye is amaranth, and it is preferably added to the test solution together with a timer solution to time the enzymatic dye-forming reaction.
