[origin: DE19757516A1] A molecule that hybridises with nucleic acids and has a backbone of linearly linked monomer units, where at least one of the monomer units is a non-nucleoside unit containing an acid function with a pKa of less than 3.0, is new. Independent claims are also included for the following: (1) a molecule of formula (IV): where L = H, OH, 1-4C alkanoyl, an optionally protected nucleobase-binding group, 6-14C aryl, N-, O- and/or S-containing heterocyclyl, or an intercalator or reporter group; A = a spacer of 1-3 atoms; x = 0-3; E and F = COOX, CSOX, SO2X, SO3X or NR<1>Y; R<1> = H or 1-3C alkyl; X = H or a protecting/activating group; Y = H or a protecting group; N = amine nitrogen; C and D = (CR<2>R<3>)n or CR<2>R<3>CR<4>R<5>; R<2>-R<5> = H, optionally substituted 1-6C alkyl, OH, COOH or an optionally protected group containing an acid function with a pKa of less than 3.0; n = 1-4; (2) a method for detecting the presence or absence of a nucleic acid in a sample suspected of containing one or more other nucleic acids, whose base sequence differs from that of the target nucleic acid in one or more positions, comprising contacting the sample with a probe whose base sequence is complementary to the target sequence but not to the base sequence of the other nucleic acids and determining if the probe has bound to its complementary base sequence and the probe is a molecule in which fewer than 80% but at least one of its bases are bonded to negatively charged backbone units; (3) use of negative charges in probes containing at least one non-nucleoside backbone unit to avoid nonspecific binding of the probes to (a) base sequences that are less than 50% complementary to the probes or (b) solid supports; (4) use of acid functions to increase the selectivity of probes in binding reactions; (5) use of molecules that hybridize with nucleic acids and have at least one base bonded to a negatively charged non-nucleoside backbone unit as primers for polymerase-catalysed extension with the aid of mononucleoside triphosphates; and (6) use of molecules that bind to nucleic acids and have at least one base bonded to a negatively charged non-nucleoside backbone unit for transfection into cells with the aid of transfection reagents.
