Abstract (en)

[origin: DE19735994A1] Enzymatic synthesis of guanosinediphosphate (GDP)-6-deoxyhexoses (I) from simple nutrients or GDP-D-mannose (II). Independent claims are also included for the following: (a) isolation and cloning of the genes manB (phosphomannomutase), manC (GDP-D-mannose synthase), gmd (GDP-D-mannose-4,6-dehydratase), rfbD (as gmd), wcaG (GDP-4-keto-6-deoxymannose-3,5-epimerase-4-reductase) or rffT (fucosyl transferase) from Escherichia coli, wbcH (fucosyl transferase) from Yersinia enterocolitica, fucT (fucosyl transferase) from Helicobacter pylori, the gene for perosaminyl tranferase from Vibrio cholerae O1 or other bacteria, in suitable cloning vectors and transformation into bacteria or fungi; (b) active expression of these genes; (c) transfer of GDP-activated sugars using glycosyl transferases.
