[origin: DE19741607A1] A synthetic polypeptide comprising at least one "defined" PrP sequences or sequences derived therefrom that are recognised by PrP(Sc)-binding substances PrP = prion protein; PrP(Sc) = disease-specific isoform of PrP. Independent claims are also included for the following: (1) a pharmaceutical composition for therapy of prion diseases, comprising at least one synthetic polypeptide as above or at least one PrP(Sc)-binding substance that recognises the "defined" sequences; (2) an agent for diagnosis of prion diseases, comprising at least one synthetic polypeptide as above or at least one PrP(Sc)-binding substance that recognises the "defined" sequences; (3) a vaccine for prevention and therapy of prion diseases, comprising at least one synthetic polypeptide as above or at least one PrP(Sc)-binding substance that recognises the "defined" sequences; (4) a DNA molecule coding at least for a synthetic polypeptide as above; (5) a process for producing PrP(Sc)-specific antibodies, comprising immunising nonhuman mammals with at least one polypeptide as above and isolating the resulting antibodies; (6) a method for detecting PrP(Sc)-specific surface sequence regions, comprising incubating a PrP-specific peptide library with PrP(Sc)-binding substances and visualising the binding regions of the peptide library; and (7) use of polypeptides as above in a pharmaceutical or chemical library for the detection of PrP(Sc)-specific agents.
