[origin: WO0007015A1] The invention provides test strip assay devices for detection of an analyte in a fluid analyte sample in which the hook effect is minimized and interference with binding reactions in the assay are substantially avoided, as well as methods for use of the devices to those ends. To these ends, the devices of the invention separate the test zone of the test strip (in which a binder for the analyte is immovably bound) and the tracer zone of the test strip (in which a reconstitutable tracer is movably bound) in horizontal and, in some embodiments, in vertical planes. In all embodiments of the devices and methods of the invention, fluid analyte sample applied to the test strip contacts the test zone before the tracer contacts either the sample or the test zone. The device is especially useful for detection of analyte whose binding with assay binders is especially sensitive to assay conditions, such as antibody analyte.
