Abstract (en)
[origin: WO9733169A1] Invented is a method for determining the affinity of protein targets for chemical agents during screening of combinatorial libraries. The libraries are synthesized on solid supports with a gradient of coupling sites, such that the target is presented with a range of compound concentrations. The distribution of protein bound to the immobilized compounds is measured by quantitative image analysis. Compounds are ranked in order of binding affinity by comparing their ability to bind across the gradient.
