Abstract (en)

[origin: DE19812103A1] Synthesis of a nucleic acid (NA) comprises: (1) preparing a first NA (1) with at least one end that permits annealing and/or linkage of another NA (2); (2) annealing/linking (2) to the end of (1), with the other end of (2) masked; (3) masking the end of NA where (2) has annealed; (4) cleaving (2) at a predetermined site to remove the mask and to generate an end that can react (as above) with further NA; and (5) (v) repeating the cycle (ii)-(iv) at least once, using any suitable NA in step (ii). An Independent claim is also included for a kit comprising at least a ligase. a polymerase, a type II S-restriction enzyme and/or uracil-DNA-glycosylase, a pyrimidase and/or endonuclease III and a formamidinopyrimidine DNA glycosylase and/or a mismatch repair enzyme. The kit may also comprise: (1) a phosphatase, (2) a terminal transferase, and/or exonuclease; and (3) a synthetic matrix, optionally with starter NA attached to it.
