Abstract (en)

[origin: DE19801780A1] Production of a transgenic non-human animal (I) is new, and comprises transfecting an embryonic stem cell line with a recombinant vector (II) (comprising a p53 knock in gene), isolating the stably transfected cell clone (III) and implanting into a female non-human animal. The production of (I) comprises transfecting an embryonic stem cell line of (I) with (II), isolating the stably transfected (III) and implanting (III) into a female, non-human animal and a descendant having the selected gene exchange maintained. The recombination event comprises replacing all/part of a p53 gene of the non-human animal by a functional homologous p53 gene or p53 gene segment of another animal, where the respective knock-in sequence is placed in the genomic sequence. Independent claims are also included for: (1) (I), where all or part of a p53 gene has been replaced by a homologous functional p53 gene or p53 gene segment of another animal; (2) a selectable marker (II) for homologous recombination in a method as above; and (3) (III).
