Abstract (en)

[origin: WO9947704A1] The invention relates to compositions (reporter systems) containing a DNA sequence coding for a ribozyme as well as an oligonucleotide substrate, which is split by the ribozyme transcribed by the DNA sequence. According to a preferred version of the invention the oligonucleotide substrate is marked with a fluorophoric group (reporter group) and a fluorescence-quenching group (quencher group). After splitting by the ribozyme, quenching of the fluorescence of the fluorophore by the fluorescence-quenching group is suppressed, so that a fluorescence signal is generated. The invention also relates to methods for measuring transcription rates, for example to determine transcription inhibitors or transcription activators.
