Abstract (en)
[origin: WO0006766A1] There is provided a method for detecting and/or differentially identifying long-term repopulating hematopoietic progenitor cells. The method comprises detecting expression of tissue transglutaminase, which is uniquely expressed by long-term repopulating cells among blood or marrow cells. Thus, the method can comprise, for example, detecting the expressed protein, detecting an mRNA encoding the protein, or detecting an enzymatic metabolite of the protein. There is also provided a method for isolating long-term repopulating hematopoietic progenitor cells, comprising contacting cells from a sample of blood or marrow with an immobilized probe that selectively binds to tissue transglutaminase, to bind the long-term repopulating cells, and then collecting the bound cells. The selective identification or isolation of such long-term repopulating hematopoietic progenitor cells provides specific advantages, including the potentiation of allograft procedures, marrow reconstitution following myeloablation, and similar serious medical procedures.
