[origin: WO0121310A2] The present invention provides, in one embodiment, a capillary cassette to facilitate rapid dialysis or thermocycling, via liquid or gas, of sub-microliter samples. In one embodiment, the invention is configured for use with a fluid handling machine for automated, high-throughput processing. Dialysis or thermocycling can be performed while the capillary cassette is mounted on the fluid handling machine. However, another embodiment of the invention provides higher throughput by the use of a hotel to simultaneously process a plurality of capillary cassettes. Hotel capabilities include dialysis, thermocycling, cleaning and chemical regeneration. Exemplary applications include but are not limited to polymerase chain reaction (PCR), DNA sequencing applications, oligonucleotide ligation, ligase chain reaction (LCR), single nucleotide extension, exonuclease treatment, and oligonucleotide hybridization assays.
