[origin: WO0125759A1] The invention relates to a method and a device for determining substances, such as e.g., DNA sequences, in a sample. As the substances to be analysed contained in the sample react with complementary agents on a reaction support, they are also excited to the extent that they emit fluorescent light using the evanescent field of an excitation light. The reaction support has a planar light waveguide on its top surface. The immobilised complementary agents are located on said light waveguide and either the excitation light or the fluorescent light excited by the evanescent field of the same is guided in the planar light waveguide on the top surface of the reaction support, the fluorescent light being conveyed to a high-sensitivity resolution detector via a projection lens.
