Abstract (en)

[origin: WO0151619A2] Disclosed are genetically modified fibroblast cells having the following characteristics: (a) A gene of the genome thereof is inactivated and/or a component of the signal transduction path is modified by means of a sequence of a modified AP-1 activity, said gene coding for a subunit of the transcription factor AP-1 and said component acting on the transcription factor AP-1. (b) The inventive fibroblast cell contains the gene of (a) in an expression vector and functionally combined with a promoter and in such a form that the active subunit is only expressed after induction of the promoter or the subunit that is coded by the gene is present in the inactive form and can be activated. The invention also relates to a method using the inventive fibroblast cell for identifying a compound that can promote, inhibit or modify the AP-1 dependent differentiation and/or proliferation of keratinocytes or for identifying a gene that takes part in the AP-1 dependent differentiation and/or proliferation of keratinocytes. The invention further relates to the use of the test compound or gene that has been identified by means of the inventive method, for treating or diagnosing diseases which are associated to a disturbed proliferation and/or differentiation of keratinocytes.
