[origin: WO0170751A1] The invention relates to the production of modified oligonucleotides and to their use for conjugation reactions. The invention further relates to reagents and to methods for producing aldehyde-modified oligonucleotides that contain aldehydes that are protected (masked) as acetals. Once said acetals are incorporated into the oligonucleotides the oligonucleotides are converted to aldehydes and are used for conjugation. The conjugation reaction can be carried out with the free oligonucleotide or with the oligonucleotide that is still immobilized on the substrate.
