[origin: WO0229396A2] The invention relates to a method and device for separating complex protein mixtures with the aid of high-resolution two-dimensional electrophoresis (2D electrophoresis, 2DE) in large gels. The invention is characterized in that the separation of the proteins in the first dimension is carried out in hollow, porous and protein-permeable materials and, in the second dimension, the porous materials are placed inside a glass cassette without being modified. Said glass cassette contains a flat gel into which the proteins migrate through the wall of the porous material during electrophoresis. The 2DE, together with the mass spectrometry, forms the basic technique, e.g. for the proteomic analysis, i.e. for separating and identifying the entire protein of a cell type, organ or of an organism.
