[origin: EP1559783A1] Method for chromatographic separation of a nucleic acid (NA) mixture so that plasmid-DNA (I) is separated from other components, especially other NA, is new. Method for chromatographic separation of a nucleic acid (NA) mixture so that plasmid-DNA (I) is separated from other components, especially other NA, comprises: (A) optionally adjusting the conductivity of the starting mixture to 70-95 mS at pH 4.8-5.4 and 20[deg]C by adding one or more alkali and/or alkaline earth salts in an aqueous solution; (B) applying the mixture to a chromatographic material; (C) washing at least once with a solution of 0.9-1.8 M alkali salt and/or 0.1-0.24 M alkaline earth salt, pH 7-7.4; and (D) eluting (I) with a solution of at least 1.3 M alkali salt or 0.27 M alkaline earth salt, pH 7-7.4.
