Abstract (en)

[origin: WO2020108948A1] A light microscope comprises a scanner (25) for scanning a specimen (35) using illumination light (12) and a light detector (60) for measuring specimen light (15). A microlens array (50), which comprises a plurality of microlenses (51-54), is arranged upstream of the light detector (60) in the region of a pupil plane. The light detector (60) comprises, downstream of each microlens (51-54), a plurality of detector elements (61-65) and has a complete readout frequency of at least 100 kHz. Wavefront information with respect to the specimen light (15) is determined by means of the detector elements (61-65) arranged downstream of each microlens (51-54). Furthermore, signals of the detector elements (61-65) are added together to form a specimen point signal. By means of the scanner (25), illumination light (12) is directed at different specimen points in succession and respective specimen point signals are captured, wavefront information also being determined for at least some of the different specimen points. The determined wavefront information can be used in a calculation of a specimen image from the plurality of specimen point signals.
