[origin: CN116249559A] The present disclosure provides methods and compositions comprising polynucleotides comprising nucleic acids encoding anti-idiotypic polypeptides, as well as polypeptides encoded by the polynucleotides and cells comprising and expressing the polypeptides. The disclosed methods include methods of utilizing anti-idiotype polypeptides as safety switches when the anti-idiotype polypeptides are used in combination with antibodies, including approved biological antibodies that include recognized idiotypes. Certain embodiments include an anti-idiotypic polypeptide and a nucleotide encoding the anti-idiotypic polypeptide, the nucleotide including an internal domain. In some embodiments, this internal domain has a functional domain that can induce proliferation or cell death upon binding of the anti-idiotypic polypeptide to its target antibody.
