Description

FIELD OF THE INVENTION



[0001] The invention is directed to a class of fused pyridone compounds, their salts, pharmaceutical compositions comprising them and their use in therapy of the human body. In particular, the invention is directed to a class of hydroxycycloalkane fused pyridone compounds, which are muscarinic M1 receptor positive allosteric modulators and hence are useful in the treatment of Alzheimer's Disease and other diseases mediated by the muscarinic M1 receptor.

BACKGROUND OF THE INVENTION



[0002] Alzheimer's Disease is a common neurodegenerative disease affecting the elderly, resulting in progressive memory impairment, loss of language and visuospatial skills, as well as behavior deficits. Characteristics of the disease include degeneration of cholinergic neurons in the cerebral cortex, hippocampus, basal forebrain, and other regions of the brain, neurofibrillary tangles, and accumulation of the amyloid β peptide (Aβ). Aβ is a 39-43 amino acid produced in the brain by processing of the beta-amyloid precursor protein (APP) by the beta-amyloid protein cleaving enzyme ("beta secretase" or "BACE") and gamma-secretase. The processing leads to accumulation of Aβ in the brain.

[0003] Cholinergic neurotransmission involves the binding of acetylcholine either to the nicotinic acetylcholine receptor (nAChR) or to the muscarinic acetylcholine receptor (mAChR). It has been hypothesized that cholinergic hypofunction contributes to the cognitive deficits of patients suffering from Alzheimer's Disease. Consequently, acetyl cholinesterase inhibitors, which inhibit acetylcholine hydrolysis, have been approved in the United States for use in the treatment of the cognitive impairments of Alzheimer's Disease patients. While acetyl cholinesterase inhibitors have provided some cognitive enhancement in Alzheimer's Disease patients, the therapy has not been shown to change the underlying disease pathology.

[0004] A second potential pharmacotherapeutic target to counteract cholinergic hypofunction is the activation of muscarinic receptors. Muscarinic receptors are prevalent throughout the body. Five distinct muscarinic receptors (M1-M5) have been identified in mammals. In the central nervous system, muscarinic receptors are involved in cognitive, behavior, sensory, motor and autonomic functions. The muscarinic M1 receptor, which is prevalent in the cerebral cortex, hippocampus and striatum, has been found to have a major role in cognitive processing and is believed to have a role in the pathophysiology of Alzheimer's Disease. See Eglen et al, TRENDS in Pharmacological Sciences, 2001, 22:8, 409-414. In addition, unlike acetyl cholinesterase inhibitors, which are known to provide only symptomatic treatment, M1 agonists also have the potential to treat the underlying disease mechanism of Alzheimer's Disease. The cholinergic hypothesis of Alzheimer's Disease is linked to both β-amyloid and hyperphosphorylated tau protein. Formation of β-amyloid may impair the coupling of the muscarinic receptor with G-proteins. Stimulation of the M1 muscarinic receptor has been shown to increase formation of the neuroprotective αAPPs fragment, thereby preventing the formation of the Aβ peptide. Thus, M1 agonists may alter APP processing and enhance αAPPs secretion. See Fisher, Jpn J Pharmacol, 2000, 84:101-112.

[0005] However, M1 ligands that have been developed and studied for Alzheimer's Disease have produced side effects common to other muscarinic receptor ligands, such as sweating, nausea and diarrhea. See Spalding et al, Mol Pharmacol, 2042, 61:6, 1297-1302. M1 ligands are disclosed for instance in WO 2008/002621 and WO 2007/100366.

[0006] The muscarinic receptors are known to contain one or more allosteric sites, which may alter the affinity with which muscarinic ligands bind to the primary binding or orthosteric sites. See, e.g., S. Lazareno et al, Mol Pharmacol, 2002, 62:6, 1491-1505; S. Lazareno et al, Mol Pharmacol, 2000, 58, 194-207.

[0007] Thus the compounds of the invention, which are muscarinic M1 receptor positive allosteric modulators, are believed to be useful in the treatment of Alzheimer's Disease and other diseases mediated by the muscarinic M1 receptor.

SUMMARY OF THE INVENTION



[0008] The present invention is novel fused pyridone compounds of generic formula (I)

or a pharmaceutically acceptable salt thereof, which is useful as an M1 receptor positive allosteric modulator.

[0009] The invention is further directed to methods of treating a patient (preferably a human) for diseases or disorders in which the M1 receptor is involved, such as Alzheimer's disease, cognitive impairment, schizophrenia, pain disorders and sleep disorders, by administering to the patient a therapeutically effective amount of a compound of general formula (I), or a pharmaceutically acceptable salt thereof. The invention is also directed to pharmaceutical compositions which include an effective amount of a compound of formula (I), or a pharmaceutically acceptable salt thereof, and a pharmaceutically acceptable carrier, and the use of the compounds and pharmaceutical compositions of the invention in the treatment of such diseases.

DETAILED DESCRIPTION OF THE INVENTION



[0010] In one embodiment, the invention is directed to compounds of general formula (I)

and pharmaceutically acceptable salts thereof, wherein
Q is selected from the group consisting of
  1. (1) -CH2-, or
  2. (2) -CH2CH2-;
R1 and R2 are each selected from the group consisting of
  1. (1) hydrogen,
  2. (2) hydroxy,
  3. (3) -C1-6 alkyl,
  4. (4) -OC1-6 alkyl,
  5. (5) -C1-3 alkyl-C6-10 aryl, or
  6. (6)-N(R7R8),
wherein any R1 or R2 alkyl or aryl moiety is optionally substituted with one or more halogen,
or R1 and R2 together form the group =O;
R3 and R4 are each selected from the group consisting of
  1. (1) hydrogen,
  2. (2) hydroxy,
  3. (3) -C1-6 alkyl, or
  4. (4)-OC1-6 alkyl,
or R3 and R4 together form the group =O;
R5 is selected from the group consisting of
  1. (1) -C6-10 aryl, and
  2. (2) -C2-4 alkenyl-C6-10 aryl,
    wherein any R5 aryl or pyridyl moiety is optionally substituted with one or more
    1. (a) halogen,
    2. (b) hydroxy,
    3. (c) cyano,
    4. (d) -(CH2)q-aryl,
    5. (e) -O-(CH2)q-aryl,
    6. (f) -C1-6 alkyl,
    7. (g) -OC1-6 alkyl,
    8. (h) heteroaryl, wherein said heteroaryl is an aromatic cyclic group having from 5 to 12 cyclic atoms, said cyclic atoms selected from carbon, nitrogen, oxygen or sulfur,
    9. (i) -N(R7R8),
      and wherein said alkyl, aryl or heteroaryl moiety is optionally substituted with one or more
      1. (i) halogen,
      2. (ii) hydroxy,
      3. (iii) cyano,
      4. (iv) -C1-6 alkyl, or
      5. (v) -OC1-6 alkyl;
R6 is selected from the group consisting of
  1. (1) hydrogen,
  2. (2) -C1-6 alkyl, and
  3. (3) -CH2-aryl,
    wherein any R6 alkyl or aryl moiety is optionally substituted with one or more
    1. (a) halogen,
    2. (b) cyano, and
    3. (c) -O-C1-6 alkyl, wherein said alkyl is optionally substituted with one or more halo;
R7 and R8 are each selected from the group consisting of
  1. (1) hydrogen,
  2. (2) -C1-6 alkyl,
  3. (3) -C1-3 alkyl-C6-10 aryl,
    and any R7 or R8 alkyl or aryl moiety is optionally substituted with one or more halogen, or R7 and R8 and the nitrogen atom to which they are both bonded are linked together to form a 5- or 6-membered ring; and
q is 0 or 1
or a compound selected from:

1-[((5-bromopyridin-2-yl)methyl]-5-hydroxy-4-oxo-1,4,5,6,7,8-hexahydroquinoline-3-carboxylic acid,

1-(((5-(3-aminophenyl)pyridin-2-yl)methyl)-5-hydroxy-4-oxo-1,4, 5,6,7,8-hexahydroquinoline-3-carboxylic acid,

1-(((5-(4-fluorophenyl)pyridin-2-yl)methyl)-5-hydroxy-4-oxo-1,4,5,6,7,8-hexahydroquinoline-3-carboxylic acid,

[0019] The invention discloses methods of treating a patient (preferably a human) for diseases in which the M1 receptor is involved, such as Alzheimer's Disease, cognitive impairment, schizophrenia, pain disorders and sleep disorders, by administering to the patient a therapeutically effective amount of a compound of general formula (I).

[0020] The invention discloses the use of a compound of formula (I) for treating diseases or disorders in which the M1 receptor is involved, such as Alzheimer's disease, cognitive impairment, schizophrenia, pain disorders and sleep disorders.

[0021] The invention is also directed to medicaments or pharmaceutical compositions for treating diseases or disorders in which the M1 receptor is involved, such as Alzheimer's disease, cognitive impairment, schizophrenia, pain disorders and sleep disorders, which comprise a compound of formula (I), or a pharmaceutically acceptable salt thereof, and a pharmaceutically acceptable carrier.

[0022] The invention is further directed to a method for the manufacture of a medicament or a composition for treating diseases or disorders in which the M1 receptor is involved, such as Alzheimer's disease, cognitive impairment, schizophrenia, pain disorders and sleep disorders, by combining a compound of formula (I) with one or more pharmaceutically acceptable carriers.

After stirring for 7 hrs at ambient temperature, the mixture was poured into water and extracted three times with ethyl acetate. The combined ethyl acetate extracts were washed once with water and brine, dried with sodium sulfate, filtered and concentrated in vacuo to afforded ethyl 1-[3-bromoprop-2-en-1-yl]-4,5-dioxo-1,4,5,6,7,8-hexahydroquinoline-3-carboxylate as a mixture of E- and Z-isomers. Additional compound was obtained by concentration of the aqueous extract in vacuo and extraction of the resulting residue with a mixture of 80% chloroform and 20% methanol (v/v).

The mixture was stirred at ambient temperature for 18 hrs, and then partitioned between water and ethyl acetate. The aqueous layer was extracted twice with ethyl acetate and the combined ethyl acetate extracts were washed with water and brine, dried over sodium sulfate, filtered and concentrated in vacuo to provide ethyl 1-(4-bromobenzyl)-4,5-dioxo-1,4,5,6,7,8-hexahydroquinoline-3-carboxylate.

Determining Activity of Putative Compounds:

1. A compound of formula (I):

Q is selected from the group consisting of

R1 and R2 are each selected from the group consisting of

(1) hydrogen,

(2) hydroxy,

(3) -C1-6 alkyl,

(4) -OC1-6 alkyl,

(5) -C1-3 alkyl-C6-10 aryl, or

(6) -N(R7R8),
wherein any R1 or R2 alkyl or aryl moiety is optionally substituted with one or more halogen,

or R1 and R2 together form the group =O;
R3 and R4 are each selected from the group consisting of

(1) hydrogen,

(3) -C1-6 alkyl, or

or R7 and R8 and the nitrogen atom to which they are both bonded are linked together to form a 5- or 6-membered ring; and

or a compound selected from:

1-[((5-bromopyridin-2-yl)methyl]-5-hydroxy-4-oxo-1,4,5,6,7,8-hexahydroquinoline-3-carboxylic acid,

1-(((5-(3-aminophenyl)pyridin-2-yl)methyl)-5-hydroxy-4-oxo-1,4,5,6,7,8-hexahydroquinoline-3-carboxylic acid,

1-(((5-(4-fluorophenyl)pyridin-2-yl)methyl)-5-hydroxy-4-oxo-1,4,5,6,7,8-hexahydroquinoline-3-carboxylic acid,

3. A compound of claim 1, or a pharmaceutically acceptable salt thereof, wherein R2 is hydrogen or methyl.
 
4. A compound of claim 1, or a pharmaceutically acceptable salt thereof, wherein R1 and R2 together form oxo.
 
5. A compound of claim 4, or a pharmaceutically acceptable salt thereof, wherein R1 and R2 together form oxo and R3 and R4 together form oxo.
 
6. A compound of claim 1, or a pharmaceutically acceptable salt thereof, wherein R1 and R2 are each selected from the group consisting of

(1) hydrogen,

(2) hydroxy,

(3) -C1-6 alkyl,

(4) -OC1-6 alkyl,

(5) -C1-3 alkyl-C6-10 aryl, or

(6) -N(R7R8),
wherein any R1 or R2 alkyl or aryl moiety is optionally substituted with one or more halogen,

7. A compound of claim 1, or a pharmaceutically acceptable salt thereof, wherein R1 is selected from the group consisting of

(1) hydrogen,

(6) -N(R7R8),

wherein any R1 alkyl or aryl moiety is optionally substituted with one or more halogen, and R2, R3 and R4 are each hydrogen.
 
8. A compound of any of claims 1 to 7, or a pharmaceutically acceptable salt thereof,
wherein R6 is hydrogen.
 
9. A compound of claim 1, wherein the compound of formula (I) is a compound of formula (II):

or a pharmaceutically acceptable salt thereof, wherein R1, R2, R3, R4 and R6 are as in claim 1, and R10 is optionally present at one or more of the phenyl ring carbon atoms, and is selected from the group consisting of

(1) halogen,

(2) hydroxy,

(3) cyano,

(4) -(CH2)q-aryl,

(5) -O-(CH2)q-aryl,

(6) -C1-6 alkyl,

(7) -OC1-6 alkyl,

(8) heteroaryl, wherein said heteroaryl is an aromatic cyclic group having from 5 to 12 cyclic atoms, said cyclic atoms selected from carbon, nitrogen, oxygen or sulfur,

(9) -N(R7R8),
and wherein said alkyl, aryl or heteroaryl moiety is optionally substituted with one or more

R1 und R2 jeweils ausgewählt sind aus der Gruppe, bestehend aus

(1) Wasserstoff,

(2) Hydroxy,

(3) -C1-6-Alkyl,

(4) -OC1-6-Alkyl,

(5) -C1-3-Alkyl-C6-10-aryl oder

(6) -N(R7R8),
wobei jeder R1- oder R2-Alkyl- oder -Arylrest gegebenenfalls substituiert ist mit einem oder mehreren Halogen,

oder R1 und R2 zusammen die Gruppe =O bilden,
R3 und R4 jeweils ausgewählt sind aus der Gruppe, bestehend aus

(1) Wasserstoff,

(h) Heteroaryl, wobei das Heteroaryl eine aromatische zyklische Gruppe mit 5 bis 12 Zyklus-Atomen ist, wobei die Zyklus-Atome ausgewählt sind aus Kohlenstoff, Stickstoff, Sauerstoff oder Schwefel,

(i) -N(R7R8),
und wobei der Alkyl-, Aryl- oder Heteroarylrest gegebenenfalls substituiert ist mit einem oder mehreren

R7 und R8 jeweils ausgewählt sind aus der Gruppe, bestehend aus

(1) Wasserstoff,

(2) -C1-6-Alkyl,

(3) -C1-3-Alkyl-C6-10-aryl,

und jeder R7- und R8-Alkyl- oder -Arylrest gegebenenfalls substituiert ist mit einem oder mehreren Halogen,
oder R7 und R8 und das Stickstoffatom, an das sie gebunden sind, miteinander verbunden sind unter Bildung eines 5- oder 6-gliedrigen Rings, und
q 0 oder 1 ist,
oder eine Verbindung, ausgewählt aus:

1-[((5-Brompyridin-2-yl)methyl]-5-hydroxy-4-oxo-1,4,5,6,7,8-hexahydrochinolin-3-carbonsäure,

1-(((5-(3-Aminophenyl)pyridin-2-yl)methyl)-5-hydroxy-4-oxo-1,4,5,6,7,8-hexahydrochinolin-3-carbonsäure,

1-(((5-(4-Fluorphenyl)pyridin-2-yl)methyl)-5-hydroxy-4-oxo-1,4,5,6,7,8-hexahydrochinolin-3-carbonsäure,

5-Hydroxy-4-oxo-1-((5-(thiophen-3-yl)pyridin-2-yl)methyl)-1,4,5,6,7,8-hexahydrochinolin-3-carbonsäure und

5-Hydroxy-1-((5-(1-methyl-1H-indol-5-yl)pyridin-2-yl)methyl)-4-oxo-1,4,5,6,7,8-hexahydrochinolin-3-carbonsäure,

2. Eine Verbindung nach Anspruch 1 oder ein pharmazeutisch annehmbares Salz davon, wobei jedes R1, R2, R3 und R4 Wasserstoff ist.
 
3. Eine Verbindung nach Anspruch 1 oder ein pharmazeutisch annehmbares Salz davon, wobei R2 Wasserstoff oder Methyl ist.
 
4. Eine Verbindung nach Anspruch 1 oder ein pharmazeutisch annehmbares Salz davon, wobei R1 und R2 zusammen Oxo bilden.
 
5. Eine Verbindung nach Anspruch 4 oder ein pharmazeutisch annehmbares Salz davon, wobei R1 und R2 zusammen Oxo bilden und R3 und R4 zusammen Oxo bilden.
 
6. Eine Verbindung nach Anspruch 1 oder ein pharmazeutisch annehmbares Salz davon, wobei R1 und R2 jeweils ausgewählt sind aus der Gruppe, bestehend aus

(1) Wasserstoff,

(2) Hydroxy,

(3) -C1-6-Alkyl,

(4) -OC1-6-Alkyl,

(5) -C1-3-Alkyl-C6-10-aryl oder

(6) -N(R7R8),
wobei jeder R1- oder R2-Alkyl- oder -Arylrest gegebenenfalls substituiert ist mit einem oder mehreren Halogen,

9. Eine Verbindung nach Anspruch 1, wobei die Verbindung der Formel (I) eine Verbindung der Formel (II) ist:

14. Eine Verbindung nach einem der Ansprüche 1-12 oder ein pharmazeutisch annehmbares Salz davon zur Verwendung bei der Behandlung einer Erkrankung oder Störung, die durch den muskarinischen M1-Rezeptor vermittelt wird, wobei die Erkrankung oder Störung ausgewählt ist aus der Gruppe, bestehend aus Alzheimer-Krankheit, Schizophrenie, Schmerzoder Schlafstörungen.

5. Composé selon la revendication 4, ou un sel pharmaceutiquement acceptable de celui-ci, dans lequel R1 et R2 ensemble forment un oxo et R3 et R4 ensemble forment un oxo.
 
6. Composé selon la revendication 1, ou un sel pharmaceutiquement acceptable de celui-ci, dans lequel R1 et R2 sont sélectionnés chacun parmi le groupe consistant en:

(1) hydrogène,

(2) hydroxy,

(3) -alkyle C1-6,

(4) -Oalkyle C1-6,

(5) -alkyle C1-3-aryle C6-10, ou

(6) -N(R7R8),
où une fraction alkyle ou aryle quelconque de R1 ou de R2 est optionnellement substituée par un ou plusieurs halogènes,
ou bien R1 et R2 ensemble forment le groupe =O ;
et R3 et R4 sont chacun un hydrogène.


 
7. Composé selon la revendication 1, ou un sel pharmaceutiquement acceptable de celui-ci, dans lequel R1 est sélectionné parmi le groupe consistant en:

(1) hydrogène,

(2) hydroxy,

(3) -alkyle C1-6,

(4) -Oalkyle C1-6,

(5) -alkyle C1-3-aryle C6-10, ou

(6) -N(R7R8),
où une fraction alkyle ou aryle quelconque de R1 est optionnellement substituée par un ou plusieurs halogènes,
et R2, R3 et R4 sont chacun un hydrogène.


 
8. Composé selon l'une quelconque des revendications 1 à 7, ou un sel pharmaceutiquement acceptable de celui-ci,
dans lequel R6 est un hydrogène.
 
9. Composé selon la revendication 1, dans lequel le composé de la formule (I) est un composé de la formule (II):

ou un sel pharmaceutiquement acceptable de celui-ci, où R1, R2, R3, R4 et R6 sont comme dans la revendication 1 et R10 est optionnellement présent sur l'un ou plusieurs des atomes de carbone du cycle phényle et est sélectionné parmi le groupe consistant en:

(1) halogène,

(2) hydroxy,

(3) cyano,

(4) -(CH2)q-aryle,

(5) -O-(CH2)q-aryle,

(6) -alkyle C1-6,

(7) -Oalkyle C1-6,

(8) hétéroaryle, où ledit hétéroaryle est un groupe cyclique aromatique ayant de 5 à 12 atomes cycliques, lesdits atomes cycliques étant sélectionnés parmi carbone, azote, oxygène ou soufre,

(9) -N(R7R8),

et où ladite fraction alkyle, aryle ou hétéroaryle est optionnellement substituée par un ou plusieurs:

(a) halogène,

(b) hydroxy,

(c) cyano,

(d) -alkyle C1-6, ou

(e) -Oalkyle C1-6.


 
10. Composé selon la revendication 1, dans lequel le composé de la formule (I) est un composé de la formule (III):

ou un sel pharmaceutiquement acceptable de celui-ci, dans lequel R1, R5 et R6 sont tels que définis dans la revendication 1.
 
11. Composé selon la revendication 1, dans lequel le composé de la formule (I) est un composé de la formule (IV):

ou un sel pharmaceutiquement acceptable de celui-ci, dans lequel R3, R4, R5 et R6 sont tels que définis dans la revendication 1.
 
12. Composé selon la revendication 1, qui est sélectionné parmi le groupe consistant en:

acide 1-(biphényl-4-ylméthyl)-4-oxo-4,5,6,7-tétrahydro-1H-cyclopenta[b]pyridine-3-carboxylique ;

acide 1[(2E)-3-(2-fluorophényl)prop-2-én-1-yl]-4,5-dioxo-1,4,5,6,7,8-hexahydroquinoline-3-carboxylique ;

acide 1[(2Z)-3-(2-fluorophényl)prop-2-én-1-yl]-4,5-dioxo-1,4,5,6,7,8-hexahydroquinoline-3-carboxylique ;

acide 1-(biphényl-4-ylméthyl)-5-hydroxy-4-oxo-1,4,5,6,7,8-hexahydroquinoline-3-carboxylique;

13. Composition pharmaceutique comprenant un composé selon l'une quelconque des revendications 1 à 12, ou un sel pharmaceutiquement acceptable de celui-ci, et un véhicule pharmaceutiquement acceptable.
 
14. Composé selon l'une quelconque des revendications 1-12, ou un sel pharmaceutiquement acceptable de celui-ci, à utiliser dans le traitement d'une maladie ou d'un trouble survenant par la médiation du récepteur muscarinique M1, où ladite maladie ou ledit trouble est sélectionné parmi le groupe consistant en maladie d'Alzheimer, schizophrénie, douleur ou troubles du sommeil.
