    Description

    Field of the Invention



    [0001] The present invention relates to the pharmaceutical field, and provides a novel crystalline form of the compound 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide, a pharmaceutical composition comprising the novel crystalline form, as well as methods of preparation and uses thereof, as further defined in the claims.

    Background of the Invention



    [0002] The compound 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide described herein, with the chemical structure of Formula A, shows KDR kinase inhibition activities. Vascular endothelial growth factor (VEGF), and its receptor VEGFR-2, also known as kinase insert domain-containing receptor (KDR), constitute an important angiogenic pathway. Studies have shown that inhibition of KDR can cause apoptosis of endothelial cells, which consequently block the angiogenesis process (Rubin M. Tuder, Chest, 2000; 117: 281). Thus, KDR inhibitors can be used for treating angiogenesis-related disorders, such as cancer, age-related macular degeneration and chronic inflammatory disease. Studies have shown that the compound 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide can be used for treating angiogenesis-related disorders, such as the treatment of cancer.



    [0003] The ability of a compound to exist in at least one crystal structure or solid state form is known as polymorphism. Many compounds may exist as polymorph crystals and those compounds may also exist in a solid amorphous state. Until polymorphism of a compound is discovered, it is highly unpredictable (1) whether a particular compound will exhibit polymorphism, (2) how to make any such unknown polymorphs, and (3) what the properties, such as stability, will be of any such unknown polymorphs. See, e.g., J. Bernstein "Polymorphism in Molecular Crystals", Oxford University Press, (2002).

    [0004] Because the properties of any solid material depend on the structure as well as on the nature of the compound itself, different solid state forms of a compound can and often do exhibit different physical and chemical properties. Differences in chemical properties can be determined through a variety of analytical techniques to be used to characterize, analyze, and compare. And those differences in chemical properties may ultimately be used to differentiate among different solid state forms that may be discovered to exist. Furthermore, differences in physical properties, such as solubility and bioavailability, of solid state forms can be important when formulating a pharmaceutical compound. As such, novel crystalline and amorphous solid state forms of pharmaceutical compounds, such as the compound of Formula A, can be important in the development of such compounds.

    [0005] The compound 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide was described in WO 2009/137797 A2 and CN 101575333 A, and the methods of preparation thereof were also described therein.

    Summary of the Invention



    [0006] After a lot of exploration and research, we have found that the compound of Formula A(6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide) can exist in different crystalline forms, and can form solvates with certain solvents. The existence of various polymorphic forms of the compound of Formula A is explored in order to determine and prepare the most appropriate form of the compound for the pharmaceutical use. Based on these studies, the present invention provides the compound of Formula A in a new crystalline form, which is designated Form I. Also disclosed herein is the compound of Formula A in further crystalline forms, solvates and the crystalline forms thereof, which are designated Form II, Form III, Form IV, Form VII, and Form VIII, respectively.
    In one aspect, the novel crystalline forms of the compound of Formula A or the solvates thereof, which are provided or disclosed herein, are crystalline, non-hygroscopic and stable.

    [0007] In particular, the present invention provides Form I of 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide, i.e. Form I of the compound of Formula A, wherein the X-ray powder diffractogram of the Form I comprises diffraction angles expressed in degrees 2-theta (2θ) at: 5.3, 10.7, 13.9, and 14.6, each of the diffraction angles having an error of about ± 0.2 degrees (2θ).

    [0008] Also disclosed herein are solvates of 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide, which are ethanol solvate, acetic acid solvate and dioxane solvate of the compound of Formula A.

    [0009] Further, disclosed herein are solvates of 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide, which are hemiethanol solvate, monoacetic acid solvate and monodioxane solvate of the compound of Formula A.

    [0010] Even further, disclosed herein is hemiethanol solvate of 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide, which is Form II of the compound of Formula A.

    [0011] Even further, disclosed herein is Form III of 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide, i.e. Form III of the compound of Formula A.

    [0012] Even further, disclosed herein is monoacetic acid solvate of 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide, which is Form IV of the compound of Formula A .

    [0013] Even further, disclosed herein is Form VII of 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide, i.e. Form VII of the compound of Formula A. Even further, disclosed herein is monodioxane solvate of 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide, which is Form VIII of the compound of Formula A.

    [0014] In another aspect, the present invention provides methods of preparation of Form I of the compound of Formula A, which are reproducible and easy in operation. Also disclosed herein are methods of preparation of Form II, Form III, Form IV, Form VII and Form VIII.

    [0015] In still another aspect, the present invention provides pharmaceutical compositions comprising an effective amount of Form I of the compound of Formula A, as described above, and at least one pharmaceutically acceptable carrier.

    [0016] The present invention further relates to Form I of the compound of Formula A for use in treating diseases associated with KDR inhibition, for example angiogenesis-related disorders, such as cancer, age-related macular degeneration and chronic inflammatory disease, in a subject in recognized need thereof.

    [0017] In particular, the invention relates to Form I for use in treating angiogenesis-related disorders. In some embodiments, said angiogenesis-related disorders are selected from cancer, age-related macular degeneration and chronic inflammatory disease.

    DETAILED DESCRIPTION OF THE INVENTION



    [0036] The present invention provides a novel crystalline form of the compound 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide, i.e. Form I, wherein the X-ray powder diffractogram of Form I comprises diffraction angles expressed in degrees 2-theta (2θ) at: 5.3, 10.7, 13.9, and 14.6, each of the diffraction angles having an error of about ± 0.2 degrees (2θ).

    [0037] Disclosed herein are also further novel crystalline forms, solvates and the crystalline forms thereof of the compound 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide.

    [0038] The novel crystalline forms that are provided or disclosed herein are crystalline, non-hygroscopic and stable.

    [0039] As described herein, the novel crystalline forms may be identified by any one or more solid state analytical methods. For example, crystalline forms described herein may be characterized according to any one or more of, e.g., X-ray powder diffraction, lattice parameters obtained from a single crystal, Fourier Transform Infrared Spectroscopy, differential scanning calorimetry curve data, and/or a thermogravimetric curve. And if characterization by one of those methods is consistent with the existence of forms described herein, it does not mean that any one of the other methods is inconsistent with that existence.

    [0040] As described herein, the novel crystalline forms may be characterized according to X-ray powder diffraction. However, it is known in the art that the intensity and/or measured peaks in the X-ray powder diffractogram of different batches of a crystalline form may vary, because of, for example, different experimental conditions and/or preferred orientations. And according to the instrument precision, the measurement error of 2θ value is at about ±0.2 2θ. However, known is the value of the relative intensity of the peaks more dependent than peak position on certain properties of the measured samples, such as crystal size in the sample, orientation effect of crystalline and purity of the analysed materials, therefore, the deviation of the peak intensity at about ± 20% or greater range may occur. But notwithstanding experimental and machine errors, and principles such as preferred orientation, one skilled in the art can find sufficient information in the XRPD data provided herein to identify Form I and any other crystalline forms disclosed herein.

    Form I

    [0047] In some embodiments, the Form I of the compound of Formula A may have an X-ray powder diffractogram as shown in Figure 1. But notwithstanding experimental and machine errors, and principles such as preferred orientation, one skilled in the art can find sufficient information in the XRPD data provided herein to identify Form I of the compound of Formula A and any other crystalline forms.

    [0048] As disclosed herein, Form I of the compound of Formula A may also be characterized according to a DSC thermogram. In particular, the Form I of the compound of Formula A may have a DSC curve as shown in Figure 2. In the DSC thermogram, the endothermic peak of the Form I of the compound of Formula A is at about 247.3-248.3°C.

    [0049] As disclosed herein, Form I of the compound of Formula A may also be characterized by thermogravimetric analysis (TGA). In particular, the Form I of the compound of Formula A may have a TGA curve as shown in Figure 3 indicating the Form I as described herein as an anhydrous material or a neat form.

    [0050] In some embodiments, Form I of the compound of Formula A is substantially free of other crystalline Forms as described herein. For example, the content of Form I of the compound of Formula A is at least 99%, at least 95%, at least 90%, or even lower to 80%, by weight. Further for example, the content of Form I of the compound of Formula A is at least 70%, or at least 60%, by weight. Even further for example, the content of Form I of the compound of Formula A is at least 50% by weight.

    Methods of Preparing Form I


    Method A



    [0051] Also provided herein is a method of preparing Form I of the compound of Formula A, comprising:
    1. (1) mixing compound 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide with at least one dissolution solvent or dissolution solvent mixture, and heating the mixture to reflux to obtain a solution;

    Example 40 (Reference): Preparation of Form IV of monoacetic acid solvate of the compound of Formula A

(DSC): 245.4-247.8°C.

    wherein the X-ray powder diffractogram of the Form I comprises diffraction angles expressed in degrees 2-theta (2θ) at: 5.3, 10.7, 13.9, and 14.6, each of the diffraction angles having an error of about ± 0.2 degrees (2θ).
     
    2. The Form I according to claim 1, wherein the X-ray powder diffractogram of the Form I comprises diffraction angles expressed in degrees 2-theta (2θ) at: 5.3, 7.3, 10.7, 13.9, 14.6, and 19.9, each of the diffraction angles having an error of about ± 0.2 degrees (2θ).
     
    3. The Form I according to claim 1, wherein the X-ray powder diffractogram of the Form I comprises diffraction angles expressed in degrees 2-theta (2θ) at: 5.3, 7.3, 10.7, 13.9, 14.6, 16.3, and 19.9, each of the diffraction angles having an error of about ± 0.2 degrees (2θ).

    6. The Form I according to claim 1, wherein the Form I may have an X-ray powder diffractogram as shown in Figure 1, each of the diffraction angles having an error of about ± 0.2 degrees (2θ).
     
    7. The Form I according to any one of claims 1 to 6, wherein the content of other crystalline forms of 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide is less than 40% by weight, preferably less than 30% by weight, more preferably less than 20% by weight, more preferably less than 10% by weight, more preferably less than 5% by weight, or even more preferably less than 1% by weight.
     
    8. A pharmaceutical composition, comprising an effective amount of the Form I as defined in any one of claims 1 to 7, and at least one pharmaceutically acceptable carrier.
     
    9. The pharmaceutical composition according to claim 8, further comprising one or more other active ingredients;
    wherein said one or more other active ingredients are preferably selected from anti-neoplastic agents;

    11. The Form I as defined in any one of claims 1 to 7 or the pharmaceutical composition according to claim 8 or 9 for use in the treatment of an angiogenesis-related disorder, wherein said angiogenesis-related disorder is preferably selected from cancer, age-related macular degeneration and chronic inflammatory disease.
     
    12. The Form I as defined in any one of claims 1 to 7 or the pharmaceutical composition according to claim 8 or 9 for use in the treatment of cancer which is selected from lung cancer, head and neck cancer, colorectal cancer, pancreatic cancer, colon cancer, breast cancer, ovarian cancer, prostate cancer, stomach cancer, kidney cancer, liver cancer, brain cancer, bone cancer, sarcoma and leukemia, preferably wherein said cancer is colorectal cancer, lung cancer, stomach cancer or kidney cancer.
     
    13. The Form I as defined in any one of claims 1 to 7 for use in the treatment of cancer,
    wherein said Form I is to be administered in combination with one or more other active ingredients,
    wherein said Form I and said one or more other active ingredients are formulated as separate compositions or are formulated in the same pharmaceutical composition,

    14. A method which is:

    (i) a method of preparing the Form I according to any one of claims 1 to 7, comprising:

    (1) mixing compound 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide with at least one dissolution solvent or dissolution solvent mixture, and heating the mixture to reflux to obtain a solution;
    wherein, said at least one dissolution solvent is selected from methanol, C3-6 alkanol, acetic acid, and aprotic solvent; said dissolution solvent mixture is selected from a mixture of two or more aprotic solvents, or a mixture of water miscible organic solvent and water, in which the volume percentage of the water miscible organic solvent in said dissolution solvent mixture is less than about 50%;

    (2) cooling the solution obtained in step (1) to ambient temperature slowly; then

    (3) isolating to obtain the solid of Form I of 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide;

    (4) drying the solid obtained in step (3); or

    (ii) a method of preparing the Form I according to any one of claims 1 to 7, comprising:

    (1) mixing compound 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide with at least one dissolution solvent, then heating the mixture to reflux to obtain the first solution; wherein said dissolution solvent is selected from ethanol, isopropanol, acetone, dichloromethane, dimethyl sulfoxide, and N,N-dimethylformamide;

    (2) adding at least one anti-dissolution solvent to said first solution to obtain the second solution;

    (3) leaving said second solution to cool spontaneously slowly to ambient temperature; then

    (4) isolating to obtain the solid of Form I of 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide;

    (5) drying the solid obtained in step (4); or

    (iii) a method of preparing the Form I according to any one of claims 1 to 7, comprising:

    (1) suspending the solid of compound 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide in an appropriate amount of a solvent system; wherein, said solvent system is selected from dissolution solvent, or a solvent mixture of water miscible organic solvent and water, in which volume percentage of water miscible organic solvent in the solvent mixture is less than about 80%;

    (2) stirring the suspension obtained in step (1) for a period of time;

    (3) isolating to obtain the solid of the Form I of 6-((6,7-dimethoxyquinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamide;

    (4) drying the solid obtained in step (3).


     
    15. The method according to claim 14, wherein the dissolution solvent used in step (1) of method (iii) is acetone.

    6. Die Form I nach Anspruch 1, wobei die Form I ein Röntgenpulverdiffraktogramm wie in Figur 1 gezeigt haben kann, wobei jeder der Beugungswinkel einen Fehler von etwa ± 0,2 Grad (2θ) aufweist.
     
    7. Die Form I nach einem der Ansprüche 1 bis 6, wobei der Gehalt an anderen kristallinen Formen von 6-((6,7-Dimethoxychinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamid weniger als 40 Gew.-%, vorzugsweise weniger als 30 Gew.-%, stärker bevorzugt weniger als 20 Gew.-%, stärker bevorzugt weniger als 10 Gew.-%, stärker bevorzugt weniger als 5 Gew.-% oder noch stärker bevorzugt weniger als 1 Gew.-% beträgt.
     
    8. Eine pharmazeutische Zusammensetzung, umfassend eine wirksame Menge der Form I, wie in einem der Ansprüche 1 bis 7 definiert, und mindestens einen pharmazeutisch verträglichen Träger.
     
    9. Die pharmazeutische Zusammensetzung nach Anspruch 8, die ferner einen oder mehrere andere Wirkstoffe enthält;
    wobei der eine oder die mehreren anderen Wirkstoffe vorzugsweise ausgewählt sind aus anti-neoplastischen Mitteln;

    10. Die Form I wie in einem der Ansprüche 1 bis 7 definiert oder die pharmazeutische Zusammensetzung nach Anspruch 8 oder 9 zur Verwendung bei der Behandlung einer Krankheit, die mit KDR-Hemmung in Zusammenhang steht.
     
    11. Die Form I wie in einem der Ansprüche 1 bis 7 definiert oder die pharmazeutische Zusammensetzung nach Anspruch 8 oder 9 zur Verwendung bei der Behandlung einer angiogenesebezogenen Erkrankung, wobei die angiogenesebezogene Erkrankung vorzugsweise aus Krebs, altersbedingter Makuladegeneration und chronischer Entzündungskrankheit ausgewählt ist.
     
    12. Die Form I wie in einem der Ansprüche 1 bis 7 definiert oder die pharmazeutische Zusammensetzung nach Anspruch 8 oder 9 zur Verwendung bei der Behandlung von Krebs, der ausgewählt ist aus Lungenkrebs, Kopf- und Halskrebs, Kolorektalkrebs, Bauchspeicheldrüsenkrebs, Dickdarmkrebs, Brustkrebs, Eierstockkrebs, Prostatakrebs, Magenkrebs, Nierenkrebs, Leberkrebs, Hirnkrebs, Knochenkrebs, Sarkom und Leukämie, wobei der Krebs bevorzugt Kolorektalkrebs, Lungenkrebs, Magenkrebs oder Nierenkrebs ist.
     
    13. Die Form I wie in einem der Ansprüche 1 bis 7 definiert zur Verwendung bei der Behandlung von Krebs,
    wobei die Form I in Kombination mit einem oder mehreren anderen Wirkstoffen zu verabreichen ist,
    wobei die Form I und der eine oder die mehreren andere/-n Wirkstoff/-e als getrennte Zusammensetzungen formuliert sind oder in derselben pharmazeutischen Zusammensetzung formuliert sind,
    wobei der eine oder die mehreren andere/-n Wirkstoff/-e bevorzugt ausgewählt sind aus anti-neoplastischen Mitteln, stärker bevorzugt aus DNA-schädigenden Chemotherapeutika, Topoisomerase I-Inhibitoren, Irinotecan, Topotecan, Camptothecin, Doxorubicin, Topoisomerase II-Inhibitoren, Etoposid, Teniposid, Daunorubicin, Alkylierungsmitteln, Melphalan, Chlorambucil, Busulfan, Thiotepa, Ifosfamid, Carmustin, Lomustin, Semustin, Streptozocin, Decarbazin, Methotrexat, Mitomycin C, Cyclophosphamid, DNA-Interkalatoren, Cisplatin, Oxaliplatin, Carboplatin, DNA-Interkalatoren und Freie-Radikalerzeugern, Bleomycin, Nukleosidmimetika, 5-Fluoruracil, Capecitibin, Gemcitabin, Fludarabin, Cytarabin, Mercaptopurin, Thioguanin, Pentostatin, Hydroxyharnstoff, die Zellreplikation störende Chemotherapeutika, Paclitaxel, Docetaxel, Vincristin, Vinblastin, Thalidomid, CC-5013, CC-4047, Protein-Tyrosinkinase-Inhibitoren, Imatinib-Mesylat, Gefitinib, Proteasom-Inhibitoren, Bortezomib, NF-kappa B-Inhibitoren, Inhibitoren der I kappa B-Kinase, Antikörper, die an Proteine binden, die bei Krebs überexprimiert werden, und dadurch die Zellreplikation herunterregulieren, Rituximab, Cetuximab und Bevacizumab,
    wobei der Krebs bevorzugt aus Lungenkrebs, Kopf- und Halskrebs, Kolorektalkrebs, Bauchspeicheldrüsenkrebs, Dickdarmkrebs, Brustkrebs, Eierstockkrebs, Prostatakrebs, Magenkrebs, Nierenkrebs, Leberkrebs, Hirnkrebs, Knochenkrebs, Sarkom und Leukämie ausgewählt ist, wobei der Krebs stärker bevorzugt aus Kolorektalkrebs, Lungenkrebs, Magenkrebs und Nierenkrebs ausgewählt ist.
     
    14. Ein Verfahren, das:

    (i) ein Verfahren zur Herstellung der Form I nach einem der Ansprüche 1 bis 7 ist, umfassend:

    (1) Mischen der Verbindung 6-((6,7-Dimethoxychinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamid mit mindestens einem Lösungsmittel oder Lösungsmittelgemisch und Erhitzen des Gemisches zum Rückfluss, um eine Lösung zu erhalten;
    wobei das mindestens eine Lösungsmittel ausgewählt ist aus Methanol, C3-6-Alkanol, Essigsäure und aprotischem Lösungsmittel; wobei das Lösungsmittelgemisch ausgewählt ist aus einem Gemisch aus zwei oder mehr aprotischen Lösungsmitteln oder einem Gemisch aus wassermischbarem organischen Lösungsmittel und Wasser, wobei der Volumenprozentsatz des wassermischbaren organischen Lösungsmittels in dem Lösungsmittelgemisch weniger als etwa 50% beträgt;

    (2) langsames Abkühlen der in Schritt (1) erhaltenen Lösung auf Umgebungstemperatur; dann

    (3) Isolieren, um den Feststoff der Form I von 6-((6,7-Dimethoxychinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamid zu erhalten;

    (4) Trocknen des in Schritt (3) erhaltenen Feststoffs; oder

    (ii) ein Verfahren zur Herstellung der Form I gemäß einem der Ansprüche 1 bis 7 ist, umfassend:

    (1) Mischen der Verbindung 6-((6,7-Dimethoxychinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamid mit mindestens einem Lösungsmittel, dann Erhitzen des Gemisches zum Rückfluss, um die erste Lösung zu erhalten; wobei das Lösungsmittel ausgewählt ist aus Ethanol, Isopropanol, Aceton, Dichlormethan, Dimethylsulfoxid und N,N-Dimethylformamid;

    (2) Hinzufügen von mindestens einem Anti-Lösungsmittel zu der ersten Lösung, um die zweite Lösung zu erhalten;

    (3) die zweite Lösung spontan langsam auf Umgebungstemperatur abkühlen lassen; dann

    (4) Isolieren, um den Feststoff der Form I von 6-((6,7-Dimethoxychinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamid zu erhalten;

    (5) Trocknen des in Schritt (4) erhaltenen Feststoffs; oder

    (iii) ein Verfahren zur Herstellung der Form I gemäß einem der Ansprüche 1 bis 7 ist, umfassend:

    (1) Suspendieren des Feststoffs der Verbindung 6-((6,7-Dimethoxychinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamid in einer geeigneten Menge eines Lösungsmittelsystems; wobei das Lösungsmittelsystem ausgewählt ist aus einem Lösungsmittel oder einem Lösungsmittelgemisch aus wassermischbarem organischen Lösungsmittel und Wasser, wobei der Volumenprozentsatz des wassermischbaren organischen Lösungsmittels in dem Lösungsmittelgemisch weniger als etwa 80% beträgt;

    (2) Rühren der in Schritt (1) erhaltenen Suspension für einen Zeitraum;

    (3) Isolieren, um den Feststoff der Form I von 6-((6,7-Dimethoxychinazolin-4-yl)oxy)-N,2-dimethylbenzofuran-3-carboxamid zu erhalten;

    (4) Trocknen des in Schritt (3) erhaltenen Feststoffs.


     
    15. Das Verfahren nach Anspruch 14, wobei das in Schritt (1) des Verfahrens (iii) verwendete Lösungsmittel Aceton ist.
     


    Revendications

    1. Forme I du 6-((6,7-diméthoxyquinazolin-4-yl)oxy)-N,2-diméthylbenzofuran-3-carboxamide, dans laquelle le diffractogramme des rayons X par la technique des poudres de la forme I comprend des angles de diffraction, exprimés en degrés 2 thêta (2θ), à : 5,3, 10,7, 13,9 et 14,6, chacun des angles de diffraction ayant une erreur d'environ ± 0,2 degré (2θ).
     
    2. Forme I selon la revendication 1, dans laquelle le diffractogramme des rayons X par la technique des poudres de la forme I comprend des angles de diffraction, exprimés en degrés 2 thêta (2θ), à : 5,3, 7,3, 10,7, 13,9, 14,6 et 19,9, chacun des angles de diffraction ayant une erreur d'environ ± 0,2 degré (2θ).
     
    3. Forme I selon la revendication 1, dans laquelle le diffractogramme des rayons X par la technique des poudres de la forme I comprend des angles de diffraction, exprimés en degrés 2 thêta (2θ), à : 5,3, 7,3, 10,7, 13,9, 14,6, 16,3 et 19,9, chacun des angles de diffraction ayant une erreur d'environ ± 0,2 degré (2θ).
     
    4. Forme I selon la revendication 1, dans laquelle le diffractogramme des rayons X par la technique des poudres de la forme I comprend des angles de diffraction, exprimés en degrés 2 thêta (2θ), à : 5,3, 7,3, 10,7, 13,9, 14,6, 16,3, 19,9, 21,1, 21,3 et 25,8, chacun des angles de diffraction ayant une erreur d'environ ± 0,2 degré (2θ).
     
    5. Forme I selon la revendication 1, dans laquelle le diffractogramme des rayons X par la technique des poudres de la forme I comprend des angles de diffraction, exprimés en degrés 2 thêta (2θ), à : 5,3, 7,3, 10,7, 13,9, 14,6, 15,2, 16,3, 19,9, 21,1, 21,3, 23,1, 23,3 et 25,8, chacun des angles de diffraction ayant une erreur d'environ ± 0,2 degré (2θ).
     
    6. Forme I selon la revendication 1, laquelle forme I peut avoir un diffractogramme des rayons X par la technique des poudres tel que présenté sur la Figure 1, chacun des angles de diffraction ayant une erreur d'environ ± 0,2 degré (2θ).
     
    7. Forme I selon l'une quelconque des revendications 1 à 6, dans laquelle la teneur en d'autres formes cristallines du 6-((6,7-diméthoxyquinazolin-4-yl)oxy)-N,2-diméthylbenzofuran-3-carboxamide est inférieure à 40 % en poids, de préférence inférieure à 30 % en poids, mieux encore inférieure à 20 % en poids, mieux encore inférieure à 10 % en poids, mieux encore inférieure à 5 % en poids, ou plus particulièrement inférieure à 1 % en poids.
     
    8. Composition pharmaceutique comprenant une quantité efficace de la forme I telle que définie dans l'une quelconque des revendications 1 à 7 et au moins un véhicule pharmaceutiquement acceptable.
     
    9. Composition pharmaceutique selon la revendication 8, comprenant en outre un ou plusieurs autres principes actifs ;
    dans laquelle lesdits un ou plusieurs autres principes actifs sont de préférence choisis parmi les agents antinéoplasiques ;
    mieux encore dans laquelle lesdits un ou plusieurs autres principes actifs sont choisis parmi les agents chimiothérapeutiques endommageant l'ADN, les inhibiteurs de topoisomérase I, l'irinotécan, le topotécan, la camptothécine, la doxorubicine, les inhibiteurs de topoisomérase II, l'étoposide, le téniposide, la daunorubicine, les agents d'alkylation, le melphalan, le chlorambucil, le busulfan, le thiotépa, l'ifosfamide, la carmustine, la lomustine, la sémustine, la streptozocine, la décarbazine, le méthotrexate, la mitomycine C, le cyclophosphamide, les agents d'intercalation d'ADN, le cisplatine, l'oxaliplatine, le carboplatine, les agents d'intercalation d'ADN et les générateurs de radicaux libres, la bléomycine, les mimétiques de nucléoside, le 5-fluorouracile, la capécitibine, la gemcitabine, la fludarabine, la cytarabine, la mercaptopurine, la thioguanine, la pentostatine, l'hydroxyurée, les agents chimiothérapeutiques qui interrompent la réplication cellulaire, le paclitaxel, le docétaxel, la vincristine, la vinblastine, le thalidomide, CC-5013, CC-4047, les inhibiteurs de protéine tyrosine kinase, le mésylate d'imatinib, le gefitinib, les inhibiteurs de protéasome, le bortézomib, les inhibiteurs de NF-kappa B, les inhibiteurs de I kappa B kinase, les anticorps qui se lient à des protéines surexprimées dans des cancers et ainsi régulent à la baisse la réplication cellulaire, le rituximab, le cétuximab, et le bévacizumab.
     
    10. Forme I telle que définie dans l'une quelconque des revendications 1 à 7 ou composition pharmaceutique selon la revendication 8 ou 9, pour une utilisation dans le traitement d'une maladie associée à une inhibition de KDR.
     
    11. Forme I telle que définie dans l'une quelconque des revendications 1 à 7 ou composition pharmaceutique selon la revendication 8 ou 9, pour une utilisation dans le traitement d'un trouble associé à une angiogenèse, où ledit trouble associé à une angiogenèse est de préférence choisi parmi un cancer, une dégénérescence maculaire liée à l'âge et une maladie inflammatoire chronique.
     
    12. Forme I telle que définie dans l'une quelconque des revendications 1 à 7 ou composition pharmaceutique selon la revendication 8 ou 9, pour une utilisation dans le traitement d'un cancer qui est choisi parmi un cancer du poumon, un cancer de la tête et du cou, un cancer colorectal, un cancer pancréatique, un cancer du côlon, un cancer du sein, un cancer ovarien, un cancer de la prostate, un cancer de l'estomac, un cancer du rein, un cancer du foie, un cancer du cerveau, un cancer des os, un sarcome et une leucémie, de préférence dans laquelle ledit cancer est un cancer colorectal, un cancer du poumon, un cancer de l'estomac ou un cancer du rein.
     
    13. Forme I telle que définie dans l'une quelconque des revendications 1 à 7 pour une utilisation dans le traitement d'un cancer,
    laquelle forme I est destinée à être administrée en combinaison avec un ou plusieurs autres principes actifs,
    laquelle forme I et lesquels un ou plusieurs autres principes actifs sont formulés sous la forme de compositions séparées ou sont formulés dans la même composition pharmaceutique,

    14. Procédé qui est :

    (i) un procédé de préparation de la forme I selon l'une quelconque des revendications 1 à 7, comprenant :

    (1) le mélange du composé 6-((6,7-diméthoxyquinazolin-4-yl)oxy)-N,2-diméthylbenzofuran-3-carboxamide avec au moins un solvant de dissolution ou un mélange solvant de dissolution, et le chauffage du mélange au reflux pour que soit obtenue une solution ;
    dans lequel ledit au moins un solvant de dissolution est choisi parmi le méthanol, un alcanol en C3 à C6, l'acide acétique, et un solvant aprotique ; ledit mélange solvant de dissolution et choisi parmi un mélange de deux solvants aprotiques ou plus, et un mélange d'eau et de solvant organique miscible avec l'eau, dans lequel le pourcentage en volume du solvant organique miscible avec l'eau dans ledit mélange solvant de dissolution est inférieur à environ 50 % ;

    (2) le lent retour à la température ambiante de la solution obtenue dans l'étape (1) ; puis

    (3) l'isolation pour que soit obtenue le solide de la forme I du 6-((6,7-diméthoxyquinazolin-4-yl)oxy)-N,2-diméthylbenzofuran-3-carboxamide ;

    (4) le séchage du solide obtenu dans l'étape (3) ; ou

    (ii) un procédé de préparation de la forme I selon l'une quelconque des revendications 1 à 7, comprenant :

    (1) le mélange du composé 6-((6,7-diméthoxyquinazolin-4-yl)oxy)-N,2-diméthylbenzofuran-3-carboxamide avec au moins un solvant de dissolution, puis le chauffage du mélange au reflux pour que soit obtenue une première solution ; dans lequel ledit solvant de dissolution est choisi parmi l'éthanol, l'isopropanol, l'acétone, le dichlorométhane, le diméthylsulfoxyde, et le N,N-diméthylformamide ;

    (2) l'addition d'au moins un anti-solvant de dissolution à ladite première solution pour que soit obtenue une deuxième solution ;

    (3) le lent retour spontané à la température ambiante de ladite deuxième solution ; puis

    (4) l'isolation pour que soit obtenue le solide de la forme I du 6-((6,7-diméthoxyquinazolin-4-yl)oxy)-N,2-diméthylbenzofuran-3-carboxamide ;

    (5) le séchage du solide obtenu dans l'étape (4) ; ou

    (iii) un procédé de préparation de la forme I selon l'une quelconque des revendications 1 à 7, comprenant :

    (1) la mise en suspension du solide du composé 6-((6,7-diméthoxyquinazolin-4-yl)oxy)-N,2-diméthylbenzofuran-3-carboxamide dans une quantité appropriée d'un système solvant ; dans lequel ledit système solvant est choisi parmi un solvant de dissolution, et un mélange solvant d'eau et de solvant organique miscible avec l'eau, dans lequel le pourcentage en volume du solvant organique miscible avec l'eau dans le mélange solvant est inférieur à environ 80 % ;

    (2) l'agitation de la suspension obtenue dans l'étape (1) pendant une certaine période de temps ;

    (3) l'isolation pour que soit obtenue le solide de la forme I du 6-((6,7-diméthoxyquinazolin-4-yl)oxy)-N,2-diméthylbenzofurane-3-carboxamide ;

    (4) le séchage du solide obtenu dans l'étape (3).


     
    15. Procédé selon la revendication 14, dans lequel le solvant de dissolution utilisé dans l'étape (1) du procédé (iii) est l'acétone.
