Description

BACKGROUND OF THE INVENTION


1. Field of the Invention



[0001] The present invention relates to a pharmaceutical composition.

2. Description of the Related Art



[0002] Fulvestrant (7α-[9-(4,4,5,5,5-pentafluoropentylsulfinyl)nonyl]estra-1,3,5(10)-triene-3,17β-diol) is an estrogen receptor antagonist and is marketed under a trade name of FASLODEX (registered trademark) as an intramuscular injection preparation by AstraZeneca. FASLODEX is supplied in the form of a 5 mL previously-filled syringe that contains fulvestrant at an indicated amount of 50 mg/mL and contains, as additives, ethanol, benzyl alcohol, benzyl benzoate, and castor oil.

[0003] In recent years, various reports on pharmaceutical preparations containing fulvestrant have been made.

[0004] For example, JP3713237B discloses a pharmaceutical preparation suitable for intramuscular injection which contains fulvestrant, pharmaceutically acceptable alcohols (ethanol and benzyl alcohol), benzyl benzoate, and castor oil.

[0005] JP2004-534093A discloses a pharmaceutical preparation suitable for intramuscular injection which contains fulvestrant, pharmaceutically acceptable alcohol (ethanol and benzyl alcohol), propylene glycol, and castor oil.

[0006] JP2009-509942A discloses a preparation that contains fulvestrant, pharmaceutically acceptable alcohol (ethanol and benzyl alcohol), propylene glycol or polyethylene glycol, and castor oil.

[0007] WO2015/033302A discloses a preparation that contains fulvestrant, benzoic acid, alcohol (ethanol), and vegetable oil.

[0008] JP2011-514349A discloses a fulvestrant blend for intramuscular injection which contains fulvestrant, glycofurol, propylene glycol, polyethylene glycol, and the like and contains substantially no castor oil. WO-2007033434A discloses a formulation comprising: 2-10% fulvestrant or salt thereof; at least one pharmaceutically acceptable alcohol; 8-15% ethanol and 5-15% benzyl alcohol, 10-20% propylene glycol and castor oil as carrier ,but no benzyl benzoate is present.

SUMMARY OF THE INVENTION



[0009] By the way, fulvestrant is a drug which shows a poor solubility in water. Therefore, in the related art, ethanol is used as a solvent in a pharmaceutical preparation containing fulvestrant.

[0010] However, use of ethanol in a pharmaceutical preparation causes the pharmaceutical preparation to have a limitation on administration to a patient who develops an allergic reaction to ethanol such as alcohol hypersensitivity, which is not preferable from the viewpoint of application to a wide range of patients.

[0011] Regarding the above-described points, the pharmaceutical preparations described in JP3713237B and JP2004-534093A contain a high concentration of ethanol in order that fulvestrant is blended at a high concentration. In addition, in the pharmaceutical preparations described in JP2009-509942A and WO2015/033302A, the preparations are made without using a non-aqueous ester solvent (benzyl benzoate or the like) in which fulvestrant has good dissolution properties, and attention is not paid to elimination of use of ethanol.

[0012] Accordingly, it is difficult to administer the pharmaceutical preparations described in JP3713237B, JP2004-534093A, JP2009-509942A, and WO2015/033302A to a patient who develops an allergic reaction to ethanol.

[0013] On the other hand, in the pharmaceutical preparation described in JP2011-514349A, the preparation is made without using ethanol. Thus, it is possible to administer the pharmaceutical preparation to a patient who develops an allergic reaction to ethanol. However, the pharmaceutical preparation described in JP2011-514349A contains substantially no castor oil. Thus, for example, in a case where intramuscular administration thereof is performed, it is difficult to maintain a therapeutically effective blood fulvestrant concentration for a certain period of time, and thus it cannot be said to be a therapeutically appropriate pharmaceutical preparation.

[0014] In addition, in selecting a solvent which is substituted for ethanol (hereinafter also referred to as "alternative solvent"), it is necessary to consider not only dissolution properties of fulvestrant in the alternative solvent but also compatibility of the alternative solvent with other ingredients to be contained (for example, a solvent such as castor oil). In particular, for an injectable liquid preparation, in a case where ingredients contained therein do not have a good compatibility, in storage at a low temperature, turbidity can occur due to phase separation or precipitation, which is not preferable.

[0015] One embodiment of the present invention has been made in view of the circumstances as described above, and an object of the present invention is to provide a pharmaceutical composition which contains substantially no ethanol, and in which fulvestrant has good dissolution properties and occurrence of turbidity is suppressed in a case of being stored at a low temperature.

[0016] Specific means for achieving the above-mentioned object includes the following embodiments.
  1. [1] A pharmaceutical composition comprising:

    fulvestrant;

    at least one kind of polyhydric alcohol selected from propylene glycol or 1,3-butylene glycol;

    benzyl alcohol;

    benzyl benzoate; and

    castor oil,

    in which the pharmaceutical composition satisfies the following requirements (1) to (6).

    1. (1) a content of ethanol is less than 1% by mass with respect to the entire mass of the pharmaceutical composition
    2. (2) a content of benzoic acid is less than 1% by mass with respect to the entire mass of the pharmaceutical composition
    3. (3) a content of the fulvestrant is 4.5% by mass to 10.0% by mass with respect to the entire mass of the pharmaceutical composition
    4. (4) a content of the at least one kind of polyhydric alcohol selected from propylene glycol or 1,3-butylene glycol is 8% by mass to 23% by mass with respect to the entire mass of the pharmaceutical composition
    5. (5) a content of benzyl alcohol is 5% by mass to 24% by mass with respect to the entire mass of the pharmaceutical composition
    6. (6) a content of benzyl benzoate is 8% by mass to 19% by mass with respect to the entire mass of the pharmaceutical composition

  2. [2] The pharmaceutical composition according to [1], in which the content of ethanol is less than 0.5% by mass with respect to the entire mass of the pharmaceutical composition.
  3. [3] The pharmaceutical composition according to [1] or [2], in which the at least one kind of polyhydric alcohol selected from propylene glycol or 1,3-butylene glycol is propylene glycol.
  4. [4] The pharmaceutical composition according to [3], in which a content of propylene glycol is 12% by mass to 18% by mass with respect to the entire mass of the pharmaceutical composition.
  5. [5] The pharmaceutical composition according to any one of [1] to [4], in which the content of benzyl alcohol is 5% by mass to 15% by mass with respect to the entire mass of the pharmaceutical composition.
  6. [6] The pharmaceutical composition according to any one of [1] to [5], in which the content of benzyl benzoate is 10% by mass to 16% by mass with respect to the entire mass of the pharmaceutical composition.
  7. [7] A pharmaceutical composition comprising:

    fulvestrant;

    propylene glycol;

    benzyl alcohol;

    benzyl benzoate; and

    castor oil,

    in which the pharmaceutical composition satisfies the following requirements (i) to (vi).

    1. (i) a content of ethanol is less than 0.5% by mass with respect to the entire mass of the pharmaceutical composition
    2. (ii) a content of benzoic acid is less than 1% by mass with respect to the entire mass of the pharmaceutical composition
    3. (iii) a content of fulvestrant is 4.5% by mass to 10.0% by mass with respect to the entire mass of the pharmaceutical composition
    4. (iv) a content of propylene glycol is 12% by mass to 18% by mass with respect to the entire mass of the pharmaceutical composition
    5. (v) a content of benzyl alcohol is 5% by mass to 15% by mass with respect to the entire mass of the pharmaceutical composition
    6. (vi) a content of benzyl benzoate is 10% by mass to 16% by mass with respect to the entire mass of the pharmaceutical composition

  8. [8] The pharmaceutical composition according to any one of [1] to [7], which is for intramuscular injection.


[0017] According to one embodiment of the present invention, there is provided a pharmaceutical composition which contains substantially no ethanol, and in which fulvestrant has good dissolution properties and occurrence of turbidity is suppressed in a case of being stored at a low temperature.

DESCRIPTION OF THE PREFERRED EMBODIMENTS



[0018] The scope of the invention is defined by the claims. Hereinafter, an example of an embodiment of a pharmaceutical composition to which the present invention is applied will be described. However, the present invention is not limited to the following embodiment at all, and it is possible to practice the present invention with appropriate modifications within a scope of a purpose of one embodiment of the present invention.

[0019] In the present specification, a numerical range expressed using "to" means a range including numerical values described before and after the preposition "to" as a minimum value and a maximum value, respectively.

[0020] In numerical ranges described in a stepwise manner in the present specification, an upper limit value or a lower limit value described in one numerical range may be replaced with an upper limit value or a lower limit value of another numerical range described in a stepwise manner. [Pharmaceutical composition]



[0025] The pharmaceutical composition of the present embodiment is a pharmaceutical composition which contains fulvestrant, at least one kind of polyhydric alcohol (hereinafter also referred to as "specific polyhydric alcohol") selected from propylene glycol or 1,3-butylene glycol, benzyl alcohol, benzyl benzoate, and castor oil, and satisfies the following requirements (1) to (6).
  1. (1) A content of ethanol is less than 1% by mass with respect to the entire mass of the pharmaceutical composition.
  2. (2) A content of benzoic acid is less than 1% by mass with respect to the entire mass of the pharmaceutical composition.
  3. (3) A content of the fulvestrant is 4.5% by mass to 10.0% by mass with respect to the entire mass of the pharmaceutical composition.
  4. (4) A content of the at least one kind of polyhydric alcohol selected from propylene glycol or 1,3-butylene glycol is 8% by mass to 23% by mass with respect to the entire mass of the pharmaceutical composition.
  5. (5) A content of benzyl alcohol is 5% by mass to 24% by mass with respect to the entire mass of the pharmaceutical composition.
  6. (6) A content of benzyl benzoate is 8% by mass to 19% by mass with respect to the entire mass of the pharmaceutical composition.


[0026] Fulvestrant, which is an active ingredient in the pharmaceutical composition of the present embodiment, is a drug which shows a poor solubility in water. Therefore, in pharmaceutical preparations containing the fulvestrant in the related art, as a solvent in which the fulvestrant has good dissolution properties, ethanol is selected.

[0027] However, use of ethanol in a pharmaceutical preparation causes the pharmaceutical preparation to have a limitation on administration to a patient who develops an allergic reaction to ethanol such as alcohol hypersensitivity, which is not preferable from the viewpoint of application to a wide range of patients.

[0028] On the other hand, in selecting an alternative solvent for ethanol, it is necessary to consider not only dissolution properties of the fulvestrant in an alternative solvent but also compatibility of the alternative solvent with other ingredients to be contained (for example, a solvent such as castor oil). In particular, for an injectable liquid preparation, in a case where ingredients contained therein do not have a good compatibility, in a case of being stored at a low temperature, turbidity can occur due to phase separation or precipitation.

[0029] The pharmaceutical composition of the present embodiment contains the fulvestrant in an amount of 4.5% by mass to 10.0% by mass with respect to the entire mass of the pharmaceutical composition, the at least one kind of polyhydric alcohol selected from propylene glycol or 1,3-butylene glycol in an amount of 8% by mass to 23% by mass with respect to the entire mass of the pharmaceutical composition, benzyl alcohol in an amount of 5% by mass to 24% by mass with respect to the entire mass of the pharmaceutical composition, benzyl benzoate in an amount of 8% by mass to 19% by mass with respect to the entire mass of the pharmaceutical composition, and castor oil, in which by setting a content of benzoic acid to be less than 1% by mass with respect to the entire mass of the pharmaceutical composition, even in a case where a content of ethanol is less than 1% by mass with respect to the entire mass of the pharmaceutical composition, that is, substantially no ethanol is contained, the fulvestrant exhibits good dissolution properties. In other words, in the pharmaceutical composition of the present embodiment, it is possible to contain the fulvestrant at a high concentration (for example, in an amount of 10.0% by mass with respect to the entire mass of the pharmaceutical composition) even in a case where substantially no ethanol is contained. In addition, in the pharmaceutical composition of the present embodiment, the above-mentioned ingredients which are contained in the pharmaceutical composition have a good compatibility. Thus, it is considered that occurrence of turbidity is suppressed even in a case of being stored at a low temperature.

<Ethanol>



[0030] In the pharmaceutical composition of the present embodiment, a content of ethanol is less than 1% by mass with respect to the entire mass of the pharmaceutical composition.

[0031] Since the content of ethanol in the pharmaceutical composition of the present embodiment is less than 1% by mass with respect to the entire mass of the pharmaceutical composition, it is possible to apply the pharmaceutical composition to even a patient who develops an allergic reaction to ethanol such as alcohol hypersensitivity. Accordingly, the pharmaceutical composition of the present embodiment can be administered to a wide range of patients including a patient who develops an allergic reaction to ethanol.

[0032] In general, in a pharmaceutical composition containing ethanol which is a volatile ingredient, there may be a case where a composition of the pharmaceutical composition changes due to volatilization of ethanol during a production step of the pharmaceutical composition, a distribution step thereof as a product, or the like. A compositional change in the pharmaceutical composition due to volatilization of ethanol causes, for example, expression of unexpected fluctuations of a clinical effect or side effects after administration, which is not preferable. In addition, as a concentration of fulvestrant in the pharmaceutical composition is increased due to volatilization of ethanol, a deviation from a quality standard may occur.

[0033] In the pharmaceutical composition of the present embodiment, a content of ethanol is less than 1% by mass with respect to the entire mass of the pharmaceutical composition. Thus, a compositional change in the pharmaceutical composition which can occur due to volatilization of ethanol during a production step or storage, in particular, a concentration change of the fulvestrant is decreased or suppressed.

[0034] Furthermore, in the pharmaceutical composition of the present embodiment, since the content of ethanol is less than 1% by mass with respect to the entire mass of the pharmaceutical composition, a burden on an operator and an environment due to volatilization of ethanol is decreased or suppressed, and an excellent production suitability is exhibited.

[0035] The content of ethanol in the pharmaceutical composition of the present embodiment is preferably less than 0.5% by mass, more preferably less than 0.3% by mass, and even more preferably less than 0.1% by mass, with respect to the entire mass of the pharmaceutical composition, and particularly preferably the pharmaceutical composition does not contain ethanol, that is, the content of ethanol is 0% by mass.

<Fulvestrant>



[0037] The pharmaceutical composition of the present embodiment contains fulvestrant as an active ingredient.

[0038] Fulvestrant (7α-[9-(4,4,5,5,5-pentafluoropentylsulfinyl)nonyl]estra-1,3,5(10)-triene-3,17β-diol) is an estrogen receptor antagonist and is known as a drug for treating breast cancer.

[0039] A content of the fulvestrant in the pharmaceutical composition of the present embodiment is 4.5% by mass to 10.0% by mass.

[0040] From the viewpoint of a single dose, the content of the fulvestrant in the pharmaceutical composition of the present embodiment is preferably 4.5% by mass to 7.0% by mass, more preferably 4.5% by mass to 5.5% by mass, and even more preferably 5% by mass to 5.5% by mass, with respect to the entire mass of the pharmaceutical composition.

<Specific polyhydric alcohol>



[0041] The pharmaceutical composition of the present embodiment contains at least one kind of polyhydric alcohol (that is, specific polyhydric alcohol) selected from propylene glycol or 1,3-butylene glycol.

[0042] In the pharmaceutical composition of the present embodiment, the specific polyhydric alcohol contributes to dissolution properties of fulvestrant.

[0043] The pharmaceutical composition of the present embodiment may contain, as the specific polyhydric alcohol, only propylene glycol, only 1,3-butylene glycol, or both of propylene glycol and 1,3-butylene glycol.

[0044] From the viewpoint of further increasing dissolution properties of fulvestrant, the pharmaceutical composition of the present embodiment preferably contains at least propylene glycol as the specific polyhydric alcohol, and it is particularly preferable that the specific polyhydric alcohol is propylene glycol.

<Benzyl alcohol>

Thus, in a case of being stored at a low temperature, there is a tendency that occurrence of turbidity due to phase separation is suppressed in the pharmaceutical composition. From the viewpoint of suppressing occurrence of such turbidity, the content of benzyl benzoate in the pharmaceutical composition of the present embodiment is preferably equal to or less than 16% by mass and more preferably equal to or less than 15% by mass, with respect to the entire mass of the pharmaceutical composition.

<Castor oil>



[0058] The pharmaceutical composition of the present embodiment contains castor oil.

[0059] Due to containing castor oil, the pharmaceutical composition of the present embodiment allows a therapeutically effective blood fulvestrant concentration to be maintained for a certain period of time.

[0060] A content of castor oil in the pharmaceutical composition of the present embodiment is not particularly limited.

[0061] From the viewpoint of maintaining dissolution properties of fulvestrant, the content of castor oil in the pharmaceutical composition of the present embodiment is, for example, preferably equal to or greater than 40% by mass, more preferably equal to or greater than 45% by mass, even more preferably equal to or greater than 50% by mass, and particularly preferably equal to or greater than 55% by mass, with respect to the entire mass of the pharmaceutical composition. 1. Dissolution properties of fulvestrant

2. Presence or absence of turbidity

The results are shown in Table 1.

Then, a mass (g) of the vial bottle in which the pharmaceutical composition was placed was measured, and the measured value was recorded.

[0129] As shown in Table 2, for the pharmaceutical compositions of Example 1, Example 3, and Examples 14 to 16 in which a content of ethanol was less than 1% by mass, even in a case of being placed in a container in an open state and left to stand at 25°C for 6 hours, no mass reduction was recognized. In addition, for the pharmaceutical compositions of Example 1, Example 3, and Examples 14 to 16, in a case of being stored at 50°C for 60 hours in a syringe-filled state, almost no mass reduction was recognized.

[0130] From these results, it was found that the pharmaceutical compositions of Example 1, Example 3, and Examples 14 to 16 exhibit a small compositional change due to volatilization of an ingredient and an excellent compositional stability.

[0131] On the other hand, the pharmaceutical compositions of Comparative Examples 13 to 29 in which a content of ethanol was equal to or greater than 1% by mass exhibit a high mass reduction rate and an inferior compositional stability, as compared with the pharmaceutical compositions of Example 1, Example 3, and Examples 14 to 16 in which a content of ethanol was less than 1% by mass.


Claims

1. A pharmaceutical composition comprising:

fulvestrant;

at least one kind of polyhydric alcohol selected from propylene glycol or 1,3-butylene glycol;

benzyl alcohol;

benzyl benzoate; and

castor oil,

wherein the pharmaceutical composition satisfies the following requirements (1) to (6).

(1) a content of ethanol is less than 1% by mass with respect to the entire mass of the pharmaceutical composition

(2) a content of benzoic acid is less than 1% by mass with respect to the entire mass of the pharmaceutical composition

(3) a content of the fulvestrant is 4.5% by mass to 10.0% by mass with respect to the entire mass of the pharmaceutical composition

(4) a content of the at least one kind of polyhydric alcohol selected from propylene glycol or 1,3-butylene glycol is 8% by mass to 23% by mass with respect to the entire mass of the pharmaceutical composition

(5) a content of benzyl alcohol is 5% by mass to 24% by mass with respect to the entire mass of the pharmaceutical composition

(6) a content of benzyl benzoate is 8% by mass to 19% by mass with respect to the entire mass of the pharmaceutical composition


 
2. The pharmaceutical composition according to claim 1,
wherein the content of ethanol is less than 0.5% by mass with respect to the entire mass of the pharmaceutical composition.
 
3. The pharmaceutical composition according to claim 1 or 2,
wherein the at least one kind of polyhydric alcohol selected from propylene glycol or 1,3-butylene glycol is propylene glycol.
 
4. The pharmaceutical composition according to claim 3,
wherein a content of propylene glycol is 12% by mass to 18% by mass with respect to the entire mass of the pharmaceutical composition.
 
5. The pharmaceutical composition according to any one of claims 1 to 4,
wherein the content of benzyl alcohol is 5% by mass to 15% by mass with respect to the entire mass of the pharmaceutical composition.
 
6. The pharmaceutical composition according to any one of claims 1 to 5,
wherein the content of benzyl benzoate is 10% by mass to 16% by mass with respect to the entire mass of the pharmaceutical composition.
 
7. A pharmaceutical composition according to claim 1 comprising:

fulvestrant;

propylene glycol;

benzyl alcohol;

benzyl benzoate; and

castor oil,

wherein the pharmaceutical composition satisfies the following requirements (i) to (vi).

(i) a content of ethanol is less than 0.5% by mass with respect to the entire mass of the pharmaceutical composition

(ii) a content of benzoic acid is less than 1% by mass with respect to the entire mass of the pharmaceutical composition

(iii) a content of fulvestrant is 4.5% by mass to 10.0% by mass with respect to the entire mass of the pharmaceutical composition

(iv) a content of propylene glycol is 12% by mass to 18% by mass with respect to the entire mass of the pharmaceutical composition

(v) a content of benzyl alcohol is 5% by mass to 15% by mass with respect to the entire mass of the pharmaceutical composition

(vi) a content of benzyl benzoate is 10% by mass to 16% by mass with respect to the entire mass of the pharmaceutical composition


 
8. The pharmaceutical composition according to any one of claims 1 to 7, which is for intramuscular injection.
 


Ansprüche

1. Pharmazeutische Zusammensetzung aufweisend:

Fulvestrant;

mindestens einen mehrwertigen Alkohol, der ausgewählt ist aus Propylenglycol oder 1,3-Butylenglycol;

Benzylalkohol;

Benzylbenzoat; und

Rizinusöl,

wobei die pharmazeutische Zusammensetzung die folgenden Anforderungen (1) bis (6) erfüllt

(1) der Gehalt an Ethanol beträgt weniger als 1 Masse-% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung

(2) der Gehalt an Benzoesäure beträgt weniger als 1 Masse-% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung

(3) der Gehalt an Fulvestrant beträgt 4,5 Masse-% bis 10,0 Masse-% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung

(4) der Gehalt an dem mindestens einen mehrwertigen Alkohol, der ausgewählt ist aus Propylenglycol oder 1,3-Butylenglycol, beträgt 8 Masse-% bis 23 Masse-% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung

(5) der Gehalt an Benzylalkohol beträgt 5 Masse-% bis 24 Masse-% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung

(6) der Gehalt an Benzylbenzoat beträgt 8 Masse-% bis 19 Masse-% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung.


 
2. Pharmazeutische Zusammensetzung nach Anspruch 1,
wobei der Gehalt an Ethanol weniger als 0,5 Masse-% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung beträgt.
 
3. Pharmazeutische Zusammensetzung nach Anspruch 1 oder 2,
wobei der mindestens eine mehrwertige Alkohol, der ausgewählt ist aus Proplyenglycol oder 1,3-Butylenglycol, Propylenglycol ist.
 
4. Pharmazeutische Zusammensetzung nach Anspruch 3,
wobei der Gehalt an Propylenglycol 12 Masse-% bis 18 Masse-% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung beträgt.
 
5. Pharmazeutische Zusammensetzung nach einem der Ansprüche 1 bis 4,
wobei der Gehalt an Benzylalkohol 5 Masse-% bis 15 Masse-% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung beträgt.
 
6. Pharmazeutische Zusammensetzung nach einem der Ansprüche 1 bis 5,
wobei der Gehalt an Benzylbenzoat 10 Masse-% bis 16 Masse-% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung beträgt.
 
7. Pharmazeutische Zusammensetzung nach Anspruch 1, aufweisend:

Fulvestrant;

Propylenglycol;

Benzylalkohol;

Benzylbenzoat; und

Rizinusöl,

wobei die pharmazeutische Zusammensetzung die folgenden Anforderungen (i) bis (vi) erfüllt

(i) der Gehalt an Ethanol beträgt weniger als 0,5 Masse-% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung

(ii) der Gehalt an Benzoesäure beträgt weniger als 1 Masse-% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung

(iii) der Gehalt an Fulvestrant beträgt 4,5 Masse-% bis 10,0 Masse-% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung

(iv) der Gehalt an Propylenglycol beträgt 12 Masse-% bis 18 Masse-% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung

(v) der Gehalt an Benzylalkohol beträgt 5 Masse-% bis 15 Masse-% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung

(vi) der Gehalt an Benzylbenzoat beträgt 10 Masse-% bis 16 Masse-% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung.


 
8. Pharmazeutische Zusammensetzung nach einem der Ansprüche 1 bis 7 zur intramuskulären Injektion.

1. Composition pharmaceutique, comprenant :

du fulvestrant ;

au moins un type de polyol sélectionné parmi le propylèneglycol ou le 1,3-butylèneglycol ;

de l'alcool benzylique ;

du benzoate de benzyle, et

de l'huile de ricin,

dans laquelle la composition pharmaceutique satisfait les exigences suivantes (1) à (6) :

(1) une teneur en éthanol est inférieure à 1 % en masse par rapport à la masse entière de la composition pharmaceutique ;

(2) une teneur en acide benzoïque est inférieure à 1 % en masse par rapport à la masse entière de la composition pharmaceutique ;

(3) une teneur en fulvestrant s'étend de 4,5 % en masse à 10,0 % en masse par rapport à la masse entière de la composition pharmaceutique ;

(4) une teneur en au moins un type de polyol sélectionné parmi le propylèneglycol ou le 1,3-butylèneglycol s'étend de 8 % en masse à 23 % en masse par rapport à la masse entière de la composition pharmaceutique ;

(5) une teneur en alcool benzylique s'étend de 5 % en masse à 24 % en masse par rapport à la masse entière de la composition pharmaceutique, et

(6) une teneur en benzoate de benzyle s'étend de 8 % en masse à 19 % en masse par rapport à la masse entière de la composition pharmaceutique.


 
2. Composition pharmaceutique selon la revendication 1,
dans laquelle la teneur en éthanol est inférieure à 0,5 % en masse par rapport à la masse entière de la composition pharmaceutique.
 
3. Composition pharmaceutique selon la revendication 1 ou 2,
dans laquelle le au moins un type de polyol sélectionné parmi le propylèneglycol ou le 1,3-butylèneglycol est le propylèneglycol.
 
4. Composition pharmaceutique selon la revendication 3,
dans laquelle une teneur en propylèneglycol s'étend de 12 % en masse à 18 % en masse par rapport à la masse entière de la composition pharmaceutique.
 
5. Composition pharmaceutique selon l'une quelconque des revendications 1 à 4,
dans laquelle la teneur en alcool benzylique s'étend de 5 % en masse à 15 % en masse par rapport à la masse entière de la composition pharmaceutique.
 
6. Composition pharmaceutique selon l'une quelconque des revendications 1 à 5,
dans laquelle la teneur en benzoate de benzyle s'étend de 10 % en masse à 16 % en masse par rapport à la masse entière de la composition pharmaceutique.
 
7. Composition pharmaceutique selon la revendication 1, comprenant :

du fulvestrant ;

du propylèneglycol ;

de l'alcool benzylique ;

du benzoate de benzyle, et

de l'huile de ricin,

dans laquelle la composition pharmaceutique satisfait les exigences suivantes (i) à (vi) :

(i) une teneur en éthanol est inférieure à 0,5 % en masse par rapport à la masse entière de la composition pharmaceutique ;

(ii) une teneur en acide benzoïque est inférieure à 1 % en masse par rapport à la masse entière de la composition pharmaceutique ;

(iii) une teneur en fulvestrant s'étend de 4,5 % en masse à 10,0 % en masse par rapport à la masse entière de la composition pharmaceutique ;

(iv) une teneur en propylèneglycol s'étend de 12 % en masse à 18 % en masse par rapport à la masse entière de la composition pharmaceutique ;

(v) une teneur en alcool benzylique s'étend de 5 % en masse à 15 % en masse par rapport à la masse entière de la composition pharmaceutique, et

(vi) une teneur en benzoate de benzyle s'étend de 10 % en masse à 16 % en masse par rapport à la masse entière de la composition pharmaceutique.


 
8. Composition pharmaceutique selon l'une quelconque des revendications 1 à 7, laquelle est destinée à une injection intramusculaire.
