    Description


    [0001] The present invention relates to novel cephem compounds and pharmaceutically acceptable salts thereof. More particularly, it relates to novel cephem compounds and pharmaceutically acceptable salts thereof, which have antimicrobial activities, to processes for preparation thereof, to pharmaceutical composition comprising the same and to the use of the novel cephem compounds and pharmaceutically acceptable salts thereof for the manufacture of medicaments for treating infectious diseases in human being and animals.

    [0002] From US-A-4 521 413 cephem compounds having an antimicrobial activity are already known which differ from the novel cephem compounds of the present invention of general formula (I) following below in that the cephem compounds of the present invention are substituted by a pyrazolio group, whereas the compounds of US-A-4 521 413 are substituted by a pyridinio group.

    [0003] In EP-A-0 047 977 cephalosporine compounds having an antibiotic activity are known which are structurally very similar to the cephem compounds (I) of the present invention, which, however, differ from the latter ones in that the pyrazolio ring of the cephem compounds of the present invention is substituted by an amino or amino substituted group, while the cephalosporine compounds of EP-A-0 047 977 do not exhibit such substituents.

    [0004] In EP-A-0 062 321 cephem compounds are disclosed having the same basic structure as the cephem compounds (I) of the present invention. The compounds of the present invention, however, can be considered to be a novel selection from the compounds disclosed in EP-A-0 062 321 in so far as (a) the heterocyclic group has been selected as being a pyrazolio group, (b) the 2-position of the pyrazolio ring is substituted by the specific group R3 and (c) a selection of further substituents on the pyrazolio ring is provided. Furthermore the cephem compounds of the present invention always have at least two substituents on the pyrazolio ring which correspond to a heterocyclic cation group of EP-A-0 062 321 and one of that plural substituents on the pyrazolio ring of the invention is always amino or substituted amino.

    [0005] Although the cephem compounds disclosed in EP-A-0 062 321 have antimicrobial activities, their antibacterial effect is no more sufficient for modern requirements.

    [0006] Accordingly, it is one object of the present invention to provide novel cephem compounds having an improved activity against pathogenic microorganisms.

    [0007] An other object of the invention is to provide processes for the preparation of these novel cephem compounds and to provide pharmaceutical compositions comprising the same as an active ingredient for treating infectious diseases caused by pathogenic microorganisms in human being and animals.

    [0008] According to one aspect the present invention relates to novel cephem compounds and pharmaceutically acceptable salts thereof which have improved antimicrobial activities and can be represented by the following general formula (I):

    R1 is amino or protected amino,

    R2 is C1-C6-alkyl or C1-C6-alkenyl,

    R3 is C1-C6-alkyl, hydroxy(C1-C6)alkyl or protected hydroxy(C1-C6)alkyl,

    R4 is amino, protected amino, Ci-C6-alkylamino,proteced Ci-C6-alkylamino, carboxy(Ci-C6)-alkylamino, N-[protected carboxy(C1-C6)alkyl]amino, and

    R7 is hydrogen or C1-C6-alkyl.



    [0009] The novel cephem compounds of the present invention have a much higher antibacterial activity than the cephem compounds disclosed in EP-A-0 062 321 as has been demonstrated by comparative tests.

    [0010] Preferred embodiments of the claimed novel cephem-compounds are described in subclaims 2 to 4.

    [0011] A particularly preferred cephem compound of the present invention is 7β-[2-(5-amino-1,2,4-thiadiazol-3-yl)-2-methoxyiminoacetamido]-3-(3-amino-2-methyl-1-pyrazolio)methyl-3-cephem-4-carboxylate (syn isomer).

    [0012] As to the novel cephem compounds of formula (I) the following points are to be noted: That is, the object compound [I] includes syn isomer, anti isomer and a mixture thereof. Syn isomer means one geometrical isomer having the partial structure represented by the following formula:

    (wherein R1 and R2 are each as defined above) and anti isomer means the other geometrical isomer having the partial structure represented by the following formula:

    (wherein R1 and R2 are each as defined above), and all of such geometrical isomers and mixture thereof are included within the scope of this invention.

    [0013] In the present specification and claim the partial structure of these geometrical isomers and mixture thereof are represented for convenient sake by the following formula:

    (wherein R1 and R2 are each as defined above).

    [0014] Another point to be noted is that the pyrazolio moiety of the compound [I] can also exist in the tautomeric form, and such tautomeric equilibrium can be represented by the following schemes.

    (wherein R3, R4 and R7 are each as defined above).

    [0015] Both of the above tautomeric isomers are included within the scope of the present invention, and in the present specification and claim, however, the object compound [I] is represented for the convenient sake by one expression of the pyrazolio group of the formula (A).

    [0016] According to a second aspect the present invention relates to a process for preparing the novel cephem compounds of general formula (I) and salts thereof, which comprises

    (1) reacting a compound of the formula :

    wherein R3, R4 and R7 are each as defined above, or its reactive derivative at the amino group or a salt thereof with a compound of the formula :

    wherein R1 and R2 are each as defined above, or its reactive derivative at the carboxy group or a salt thereof to give a compound of the formula :

    (2) subjecting a compound of the formula :

    [0017] According to a third aspect the present invention relates to a pharmaceutical composition which comprises as an active ingredient a cephem compound of formula (I) or a pharmaceutically acceptable salt thereof in admixture with pharmaceutically acceptable carriers.

    [0018] Furthermore, the present invention relates to the use of the novel cephem compounds of formula (I) or pharmaceutically acceptable salts thereof for the manufacture of medicaments for treating infectious diseases caused by pathogenic microorganisms in human being and animals.

    [0034] Suitable pharmaceutically acceptable salts of the object compound [I] are conventional non-toxic salts and include a metal salt such as an alkali metal salt [e.g. sodium salt, and potassium salt and an alkaline earth metal salt [e.g. calcium salt, and magnesium salt,], an ammonium salt, an organic base salt [e.g. trimethylamine salt, triethylamine salt, pyridine salt, picoline salt, dicyclohexylamine salt, and N,N'-dibenzylethylenediamine salt], an organic acid salt [e.g. formate, acetate, trifluoroacetate, maleate, tartrate, methanesulfonate, benzenesulfonate, and toluenesulfonate,], an inorganic acid salt [e.g. hydrochloride, hydrobromide, sulfate, and phosphate], a salt with an amino acid [e.g. arginine salt, aspartic acid salt, and glutamic acid salt].

    Process 1



    [0038] The compound [I] or a salt thereof can be prepared by reacting the compound [II] or its reactive derivative at the amino group or a salt thereof with the compound [III] or its reactive derivative at the carboxy group or a salt thereof.

    [0039] Suitable reactive derivative at the amino group of the compound [II] may include Schiff's base type imino or its tautomeric enamine type isomer formed by the reaction of the compound [II] with a carbonyl compound such as aldehyde, or ketone; a silyl derivative formed by the reaction of the compound [II] with a silyl compound such as bis(trimethylsilyl)acetamide, mono(trimethylsilyl)acetamide [e.g. N-(trimethylsilyl)-acetamide], or bis(trimethylsilyl)urea;
    a derivative formed by reaction of the compound [II] with phosphorus trichloride or phosgene.

    [0040] Suitable salts of the compound [II] and its reactive derivative can be referred to the ones as exemplified for the compound [I].

    [0041] Suitable reactive derivative at the carboxy group of the compound [III] may include an acid halide, an acid anhydride, an activated amide, and an activated ester. Suitable examples of the reactive derivatives may be an acid chloride; an acid azide; a mixed acid anhydride with an acid such as substituted phosphoric acid [e.g. dialkylphosphoric acid, phenyl phosphoric acid, diphenylphosphoric acid, dibenzylphosphoric acid, and halogenated phosphoric acid,], dialkylphosphorous acid, sulfurous acid, thiosulfuric acid, sulfuric acid, sulfonic acid [e.g. methanesulfonic acid], aliphatic carboxylic acid [e.g. acetic acid, propionic acid, butyric acid, isobutyric acid, pivalic acid, pentanoic acid, isopentanoic acid, 2-ethylbutyric acid, and trichloroacetic acid] or aromatic carboxylic acid [e.g. benzoic acid]; a symmetrical acid anhydride; an activated amide with imidazole, 4-substituted imidazole, dimethylpyrazole, triazole or tetrazole; or an activated ester [e.g. cyanomethyl ester, methoxymethyl ester, dimethyliminomethyl [(CH3)2 N = CH-] ester, vinyl ester, propargyl ester, p-nitrophenyl ester, 2,4-dinitrophenyl ester, trichlorophenyl ester, pentachlorophenyl ester, mesyl- phenyl ester, phenylazophenyl ester, phenyl thioester, p-nitrophenyl thioester, p-cresyl thioester, carboxymethyl thioester, pyranyl ester, pyridyl ester, piperidyl ester, and 8-quinolyl thioester], or an ester with a N-hydroxy compound [e.g. N,N-dimethylhydroxylamine, 1-hydroxy-2-(1 H)-pyridone, N-hydroxysuccinimide, N-hydroxyphthalimide, and 1-hydroxy-1 H-benzotriazole]. These reactive derivatives can optionally be selected from them according to the kind of the compound [III] to be used.

    [0042] Suitable salts of the compound [III] and its reactive derivative can be referred to the ones as exemplified for the compound [I].

    [0043] The reaction is usually carried out in a conventional solvent such as water, alcohol [e.g. methanol, and ethanol,], acetone, dioxane, acetonitrile, chloroform, methylene chloride, ethylene chloride, tetrahydrofuran, ethyl acetate, N,N-dimethylformamide, pyridine or any other organic solvent which does not adversely influence the reaction. These conventional solvent may also be used in a mixture with water.

    [0044] In this reaction, when the compound [III] is used in a free acid form or its salt form, the reaction is preferably carried out in the presence of a conventional condensing agent such as N,N'-dicyclohexylcarbodiimide; N-cyclohexyl-N'-morpholinoethylcarbodiimide; N-cyclohexyl-N'-(4-diethylaminocyclohexyl)-carbodiimide; N,N'-diethylcarbodiimide, N,N'-diisopropylcarbodiimide; N-ethyl-N'-(3-dimethylaminopropyl)-carbodiimide; N,N'-carbonylbis-(2-methylimidazole); pentamethyleneketene-N-cyclohexylimine; diphenylketene-N-cyclohexylimine; ethoxyacetylene; 1-alkoxy-1-chloroethylene; trialkyl phosphite; ethyl polyphosphate; isopropyl polyphosphate; phosphorus oxychloride (phosphoryl chloride); phosphorus trichloride; thionyl chloride; oxalyl chloride; Ci-C6 alkyl haloformate [e.g. ethyl chloroformate, and isopropyl chloroformate]; triphenylphosphine; 2-ethyl-7-hydroxybenzisoxazolium salt; 2-ethyl-5-(m-sulfophenyl)-isoxazolium hydroxide intramolecular salt; 1-(p-chlorobenzenesulfonyloxy)-6-chloro-1H-benzotriazole; so- called Vilsmeier reagent prepared by the reaction of N,N-dimethylformamide with thionyl chloride, phosgene, trichloromethyl chloroformate, and phosphorus oxychloride,.

    [0045] The reaction may also be carried out in the presence of an inorganic or organic base such as an alkali metal bicarbonate, tri(lower)alkylamine, pyridine, N-(C1-6)alkylmorpholine, or N,N-di(C1-6)alkylbenzylamine.

    [0046] The reaction temperature is not critical, and the reaction is usually carried out under cooling to warming.

    Process 2



    [0047] The compound [Ib] or a salt thereof can be prepared by subjecting the compound [la] or a salt thereof to elimination reaction of the carboxy protective group.

    [0048] Suitable method of this elimination reaction may include conventional one such as hydrolysis, and reduction.

    (i) For Hydrolysis :



    [0049] The hydrolysis is preferably carried out in the presence of a base or an acid including Lewis acid. Suitable base may include an inorganic base and an organic base such as an alkali metal [e.g. sodium, and potassium], an alkaline earth metal [e.g. magnesium, and calcium], the hydroxide or carbonate or bicarbonate thereof, trialkylamine [e.g. trimethylamine, and triethylamine,], picoline, 1,5-diazabicyclo[4.3.0]-non-5-ene, 1,4-diazabicyclo[2.2.2]octane, or 1,8-diazabicyclo[5.4.0]undec-7-ene.

    [0050] Suitable acid may include an organic acid [e.g. formic acid, acetic acid, propionic acid, trichloroacetic acid, and trifluoroacetic acid] and an inorganic acid [e.g. hydrochloric acid, hydrobromic acid, sulfuric acid, hydrogen chloride, and hydrogen bromide].
    The elimination using Lewis acid such as trihaloacetic acid [e.g. trichloroacetic acid, and trifluoroacetic acid, ] is preferably carried out in the presence of cation trapping agents [e.g. anisole, and phenol].

    [0051] The reaction is usually carried out in a solvent such as water, an alcohol [e.g. methanol, and ethanol], methylene chloride, tetrahydrofuran, a mixture thereof or any other solvent which does not adversely influence the reaction. A liquid base or acid can be also used as the solvent. The reaction temperature is not critical and the reaction is usually carried out under cooling to warming.

    (ii) For Reduction :



    [0052] Reduction is carried out in a conventional manner, including chemical reduction and catalytic reduction.

    [0053] Suitable reducing agents to be used in chemical reduction are a combination of a metal (e.g. tin, zinc, and iron) or metallic compound (e.g. chromium chloride, and chromium acetate) and an organic or inorganic acid (e.g. formic acid, acetic acid, propionic acid, trifluoroacetic acid, p-toluenesulfonic acid, hydrochloric acid, and hydrobromic acid).

    [0054] Suitable catalysts to be used in catalytic reduction are conventional ones such as platinum catalysts (e.g. platinum plate, spongy platinum, platinum black, colloidal platinum, platinum oxide, and platinum wire), palladium catalysts (e.g. spongy palladium, palladium black, palladium oxide, palladium on carbon, colloidal palladium, palladium on barium sulfate, and palladium on barium carbonate), nickel catalysts (e.g. reduced nickel, nickel oxide, and Raney nickel), cobalt catalysts (e.g. reduced cobalt and Raney cobalt), iron catalysts (e.g. reduced iron, and Raney iron), copper catalysts (e.g. reduced copper Raney copper, and Ullman copper. After being stirred at the same temperature for 3 hours, the mixture was added dropwise to ethyl acetate (500 ml).     Example 1



    [0103] To a solution of 7β-amino-3-[3-amino-2-(2-hydroxyethyl)-1-pyrazolio]methyl-3-cephem-4-carboxylate trihydrochloride (2 g) and N-(trimethylsilyl)acetamide (5.85 g) in tetrahydrofuran (40 ml) was added (Z)-2-(5-amino-1,2,4-thiadiazol-3-yl)-2-allyloxyiminoacetyl chloride hydrochloride (1.26 g) under ice-cooling. After being stirred for 1 hour at the same temperature, the reaction mixture was poured into diisopropyl ether (200 ml). The resulting precipitate was collected by filtration, dissolved in water (100 ml) and adjusted to pH 2.0 with 5% sodium bicarbonate solution. This solution was subjected to column chromatography on macroporous non-ionic adsorption resin "Diaion HP-20" (Trademark : prepared by Mitsubishi Chemical Industries). The desired compound was eluted with 10% isopropyl alcohol and lyophilized to give 7β-[2-(5-amino-1,2,4-thiadiazol-3-yl)-2-allyloxyiminoacetamido]-3-[3-amino-2-(2-hydroxyethyl)-1-pyrazolio]methyl-3-cephem-4-carboxylate (syn isomer) (1.07 g).

    Example 3



    [0107] To a solution of di(trifluoroacetic acid) salt of 7β-amino-3-[3-acetamido-2-(2-acetoxyethyl)-1-pyrazolio]-methyl-3-cephem-4-carboxylate (1 g) and N-(trimethylsilyl)acetamide (2.01 g) in dichloromethane (20 ml) was added (Z)-2-(5-amino-1,2,4-thiadiazol-3-yl)-2-allyloxyiminoacetyl chloride hydrochloride (0.520 g) under stirring and ice-cooling. The mixture was stirred for one hour at room temperature. The reaction mixture was added to ether under stirring and ice-cooling. The produced amorphous solid was dried in vacuo and dissolved in water. The aqueous solution was adjusted to pH 13 with 1 N aqueous sodium hydroxide under stirring at -3 - 0 ° C and stirred for 2 hours at the same temperature. The aqueous solution was adjusted to pH 2 with 1 N hydrochloric acid and subjected to column chromatography on Diaion HP-20 and the elution was carried out with 10% aqueous isopropyl alcohol.

The reaction mixture was poured into ethyl acetate (140 ml) and the resultant powder was collected by filtration. The powder was dissolved in ice- water and adjusted to pH 13 with 1 N aqueous sodium hydroxide. After stirring for 20 minutes under ice-cooling, the solution was adjusted to pH 2 with 3N hydrochloric acid and subjected to column chromatography on "Diaion HP-20" using 5% aqueous isopropyl alcohol as an eluent. Fractions containing the object compound were combined, evaporated in vacuo to remove isopropyl alcohol and lyophilized to give 7β-[2-(5-amino-1,2,4-thiadiazol-3-yl)-2-methoxyiminoacetamido]-3-(3-carboxymethylamino-2-methyl-1-pyrazolio)-methyl-3-cephem-4-carboxylate (syn isomer) (0.24 g).

    6. Verfahren zur Herstellung einer Verbindung der Formel:
